PTOX-dependent safety valve does not oxidize P700 during photosynthetic induction in the Arabidopsis pgr5 mutant

Qi Zhou et al.

Summary: The introduction of the CrPTOX2 gene from Chlamydomonas reinhardtii into Arabidopsis had mild effects on photosynthetic electron transport in the wild-type plants but partially complemented the induction of nonphotochemical quenching in the mutant plants defective in cyclic electron transport. This study suggests a reassessment of the role of PGR5 in accepting electrons and its contribution to the photoprotection of PSI during photosynthesis induction, highlighting the importance of PGR5 in oxidizing photosystem I.

An ancient function of PGR5 in iron delivery?

Dario Leister et al.

Summary: The photosynthetic apparatus in phototrophic organisms needs protection from light-induced damage. An important mechanism in plants that mitigates photo-damage is antimycin A (AA)-sensitive cyclic electron flow (CEF), whose evolution remains unclear. Researchers have found that proton gradient regulation 5 (PGR5), a key protein involved in AA-sensitive CEF, has similarities in sequence and structure with a group of ferritin-like proteins. They propose that PGR5 may have originally been involved in prokaryotic iron mobilization and delivery, which unintentionally facilitated a primordial type of CEF. The abandonment of the bacterioferritin system during the transformation of cyanobacterial endosymbionts into chloroplasts may have allowed PGR5 to specialize in CEF.

Function and Regulation of Chloroplast Peroxiredoxin IIE

Anna Dreyer et al.

Summary: Peroxiredoxins play a crucial role in redox regulation in the cell, with PRX-IIE in chloroplast protein network being found to undergo various posttranslational modifications. Its interaction with 14-3-3 upsilon protein under oxidizing conditions may guide PRX-IIE to specific target proteins for redox regulation, offering a new perspective on the cellular redox regulatory network.

In-vivo quantification of electron flow through photosystem I - Cyclic electron transport makes up about 35% in a cyanobacterium

Marius L. Theune et al.

Summary: This study presents a new method that allows quantification of electron flow through photosystem I in a photosynthetic organism in absolute terms. By specific in-vivo protocols, the redox states of various components in PSI were discerned, and the validity of the method was confirmed using parallel electron counting experiments. The study suggests that the contribution of cyclic electron transport is twice as high as assumed for plants.

The molecular pH-response mechanism of the plant light-stress sensor PsbS

Maithili Krishnan-Schmieden et al.

Summary: Research reveals that Photosystem II subunit S (PsbS) senses thylakoid lumen acidification when plants are exposed to excess light. The study utilizes NMR and IR spectroscopy to demonstrate that low pH causes repositioning of an amphipathic helix and folding of a loop involving critical pH sensing glutamate residues in PsbS.

Summary: Photosynthesis is regulated to withstand dynamic light environments, with NPQ dissipating excess excitation energy under high light intensities. There is a gap between in vitro and in vivo studies on the molecular mechanism of NPQ, and the development of a proteoliposome system provides insights into the influence of factors like PsbS on the quenching mechanism in LHCII.
