Somatic embryo initiation by rice BABY BOOM1 involves activation of zygote-expressed auxin biosynthesis genes

Plant regeneration from protoplasts of Pastinaca sativa L. via somatic embryogenesis

The isolation efficiency of protoplasts from source material reached an average of 3.6 x 10(6) cells per g of fresh mass using a mixture of cellulase and pectolyase enzymes. The regenerated plants showed morphological similarity to their donors and no change in ploidy level.

Leaf transformation for efficient random integration and targeted genome modification in maize and sorghum

Traditional transformation methods in grass species have been limited to immature embryos, hindering the widespread use of genome editing for crop improvement. Leaf transformation using Wus2 and Bbm has been successful for Cas9-mediated mutagenesis, but more complex genome editing applications require screening of a large number of regenerated plants. This study demonstrates that enhanced Wus2/Bbm expression significantly improves leaf transformation in maize and sorghum, and can be used for gene dropouts and targeted gene insertion. Furthermore, it provides a potential universal method for transformation and genome editing across the Poaceae family.

A competing endogenous RNA (ceRNA) network of DE circRNAs, DE mRNAs, DE long noncoding RNAs (lncRNAs), and DE microRNAs (miRNAs) was constructed, and the putative targets of the noncoding RNAs were significantly enriched in the KEGG pathways 'mitogen-activated protein kinase signaling' and 'nitrogen metabolism'. Furthermore, the expression patterns of the candidate circRNAs, lncRNAs, miRNAs, and mRNAs confirmed the negative correlation between miRNAs and ceRNAs.
