Functional interdependence of N6-methyladenosine methyltransferase complex subunits in Arabidopsis

Lisha Shen

Summary: Maintaining protein homeostasis among different subunits of the m(6)A methyltransferase complex is crucial for ensuring proper m(6)A deposition in plants. Two accessory subunits, FIP37 and VIR, are found to be indispensable for stabilizing the methyltransferases MTA and MTB, thus maintaining the functionality of the m(6)A methyltransferase complex. These findings reveal the interdependence among individual components in the Arabidopsis m(6)A methyltransferase complex at the post-translational level, highlighting the importance of protein homeostasis for the proper function of the complex.

Direct epitranscriptomic regulation of mammalian translation initiation through N4-acetylcytidine

Daniel Arango et al.

Summary: mRNA function is affected by modifications that can alter the behavior of nucleobases. A specific modification can have different impacts on mRNA translation depending on its position. The acetylation of cytidine (ac4C) within protein-coding sequences enhances translation, while ac4C within 5' UTRs affects protein synthesis at the initiation level. The acetylation promotes initiation at upstream sequences and competitively inhibits annotated start codons. Additionally, ac4C within AUG-flanking Kozak sequences directly impedes initiation at optimal AUG contexts.

Initiation of cytosolic plant purine nucleotide catabolism involves a monospecific xanthosine monophosphate phosphatase

Katharina J. Heinemann et al.

Summary: The study focuses on the dephosphorylation of XMP in plant cells and identifies an XMP phosphatase in Arabidopsis with specificity to XMP. The researchers demonstrate that XMPP initiates the catabolism of adenylate-derived nucleotides in vivo, while guanylates enter catabolism through a different pathway. The crystal structure and mutational analysis of XMPP provide insights into its high substrate specificity and efficient catalysis of the small XMP pool in vivo.

Cytokinin regulates vegetative phase change in Arabidopsis thaliana through the miR172/TOE1-TOE2 module

Soren Werner et al.

Summary: Cytokinin regulates the transition from juvenile to adult phase in plants by promoting the abundance of miR172, which then represses the activity of transcriptional regulators TOE1 and TOE2. The antagonistic activities of miR156 and miR172 play a crucial role in the juvenile-to-adult phase transition during plant growth.

The epitranscriptome beyond m6A

David Wiener et al.

Summary: This Perspective reviews efforts to map six different RNA modifications and how they differ from N-6-methyladenosine. The authors discuss technical and analytical challenges of characterizing the epitranscriptome and provide their conclusions on the abundance and distribution of these modifications. The article highlights commonalities and differences among the modifications in comparison to m(6)A, speculating on their origins and functional abilities over evolutionary timescales.
