Topographic representation of current and future threats in the mouse nociceptive amygdala

Summary: Adaptive behavior requires the use of both memories and ongoing experience. Amygdala neurons in mice encode sensory information to contribute to appropriate behaviors in response to both predicted and ongoing threats. The central amygdala (CeA), specifically the Calcrl-expressing population, receives spatially defined representations of internal and external stimuli, with different regions of the CeA responding to different types of threats and promoting different behaviors. The rostral CeA neurons promote locomotor responses to externally sourced threats, while the caudal CeA neurons are activated by internal threats and promote passive coping behaviors and associative valence coding. During associative fear learning, rostral CeA neurons stably encode the occurrence of noxious stimuli, while caudal CeA neurons acquire predictive responses, allowing for valence-aligned representations of current and future threats for the generation of adaptive behaviors.

The administration of cannabidiol (CBD) was able to rescue these effects, suggesting its potential therapeutic value in alcohol- and stress-related harms.

Adolescent binge-like alcohol exposure dysregulates NPY and CGRP in rats: Behavioural and immunochemical evidence

Summary: Alcohol binge drinking during adolescence has negative impacts on affective behaviour and the development of neural substrates processing affective states. This study simulated adolescent binge-like alcohol exposure in male rats and examined its effects on various affective behaviours and neuropeptide levels in functionally relevant brain regions. The results showed that binge-like alcohol exposure during adolescence led to decreased weight gain, social preference, and active stress coping, as well as dysregulated neuropeptide levels in different brain regions. These findings suggest that neuropeptide-targeted interventions may be promising for addressing negative affect during prolonged withdrawal in young individuals.
