Gegen Qinlian decoction activates AhR/IL-22 to repair intestinal barrier by modulating gut microbiota-related tryptophan metabolism in ulcerative colitis mice

Xiaojing Wang et al.

Summary: In this study, the protective effects of Gegen Qinlian decoction (GQD) on intestinal barrier damage in acute ulcerative colitis (UC) were investigated. The results showed that GQD treatment significantly alleviated the pathological symptoms and reduced bacterial infiltration in MLNs and colon tissues. GQD also repaired the structure of epithelial tight junctions, increased the expression of tight junction proteins, and regulated the diversity of gut microbiota and tryptophan metabolism.

Baicalein ameliorates ulcerative colitis by improving intestinal epithelial barrier via AhR/IL-22 pathway in ILC3s

Yan-yang Li et al.

Summary: This study elucidates the molecular mechanisms underlying the protective effect of baicalein on intestinal barrier function in colitis mice, demonstrating that baicalein improves gut inflammation, reduces intestinal permeability, and restores tight junctions possibly via promoting AhR/IL-22 pathway. Co-administration of AhR antagonist CH223191 partially blocks the therapeutic effects of baicalein, while AhR agonist FICZ ameliorates symptoms and gut barrier function in colitis mice.

Biological Activities Underlying the Therapeutic Effect of Quercetin on Inflammatory Bowel Disease

Yong-Li Lyu et al.

Summary: Inflammatory bowel disease (IBD) is a chronic autoimmune disorder characterized by immune activation and destruction of colon tissue. Quercetin, a naturally occurring compound, has the potential to treat IBD through strengthening the intestinal mucosal barrier, modulating the colonic microbiota, restoring immune homeostasis, and reducing oxidative stress. Therefore, quercetin is considered as a promising candidate for IBD treatment.

Prenylated xanthones from mangosteen (Garcinia mangostana) activate the AhR and Nrf2 pathways and protect intestinal barrier integrity in HT-29 cells

Restituto Tocmo et al.

Summary: Xanthones from mangosteen exhibit anti-inflammatory and anti-oxidative effects. The study isolated seven prenylated xanthones from mangosteen fruit pericarp extracts, with Garcinone D (GarD) showing dual protection against intestinal epithelial barrier dysfunction through AhR and Nrf2 pathways.

Indol-3-Carbinol and Quercetin Ameliorate Chronic DSS-Induced Colitis in C57BL/6 Mice by AhR-Mediated Anti-Inflammatory Mechanisms

Sina Riemschneider et al.

Summary: The study shows that quercetin and indol-3-carbinol significantly improved clinical symptoms in moderate DSS colitis, reduced histopathological scores, restored epithelial integrity, and reduced gut inflammation by lowering neutrophil and macrophage numbers. Additionally, they decreased Th17 cells while increasing Treg cells, indicating potential as promising therapeutics in IBD treatment.

AHR in the intestinal microenvironment: safeguarding barrier function

Brigitta Stockinger et al.

Summary: The Aryl hydrocarbon receptor (AHR) signaling pathway plays significant roles in the intestine, regulating various cell types and impacting inflammation and tumorigenesis. AHR is a ligand-dependent transcription factor that translocates to the nucleus upon ligand binding, where it interacts with other proteins to modulate gene expression. Research has shown that AHR not only responds to environmental pollutants, but also serves important beneficial functions in immune, epithelial, endothelial, and neuronal cells within the intestinal microenvironment.

Rules of Engagement: Epithelial-Microbe Interactions and Inflammatory Bowel Disease

Albert E. Jergens et al.

Summary: Inflammatory bowel diseases (IBD) are complex, multifactorial disorders involving a combination of environmental, genetic, dietary, mucosal immunity, and gut microbiome factors. Gut microbiota plays a pivotal role in IBD pathogenesis, while a dysfunctional intestinal epithelial barrier is also recognized as a key pathogenic factor.
