Characterization of a small molecule inhibitor of disulfide reductases that induces oxidative stress and lethality in lung cancer cells

Fraser D. Johnson et al.

Summary: Phenotype-based screening led to the identification of a compound called LCS3 that selectively impairs the growth of LUAD cells, induces oxidative stress, and inhibits the activity of disulfide reductases GSR and TXNRD1. The study also reveals that LCS3-sensitive LUAD cells are sensitive to the synergistic inhibition of glutathione and thioredoxin pathways. Furthermore, NQO1 loss is identified as a mechanism of LCS3 resistance through a genome-wide CRISPR knockout screen.

Exosome-liposome hybrid nanoparticle codelivery of TP and miR497 conspicuously overcomes chemoresistant ovarian cancer

Longxia Li et al.

Summary: This study investigated the combined application of miR497 and TP in overcoming cisplatin-resistant ovarian cancer (OC) by suppressing the mTOR signaling pathway. Bioinspired hybrid nanoparticles were prepared to deliver miR497 and TP, showing efficient uptake by tumor cells and significant anticancer activity in vitro and in vivo. Mechanistically, the nanoparticles promoted dephosphorylation of the overactivated PI3K/AKT/mTOR signaling pathway and enhanced reactive oxygen species (ROS) generation and macrophage polarization. Overall, this study offers a translational strategy to overcome cisplatin-resistant OC and potentially treat other cisplatin-resistant tumors.

Matrix Thermal Shift Assay for Fast Construction of Multidimensional Ligand-Target Space

Chengfei Ruan et al.

Summary: This article presents a new method called matrix thermal shift assay (mTSA) for fast and accurate deconvolution of ligand-binding targets and binding affinities at the proteome level. Compared to previous thermal shift assays, mTSA significantly improves analysis throughput while reducing costs and efforts. The method allows for simultaneous computation of statistical significance and fitting of dose-response profiles, resulting in more accurate identification of target proteins and reporting of binding affinities. Experimental results demonstrate the high screening sensitivity of mTSA and its potential for discovering potential targets.

Interaction of lncRNA MIR100HG with hnRNPA2B1 facilitates m6A-dependent stabilization of TCF7L2 mRNA and colorectal cancer progression

Hao Liu et al.

Summary: The study reveals that MIR100HG serves as a positive regulator of EMT in CRC, sustaining cetuximab resistance and promoting invasion and metastasis. It interacts with hnRNPA2B1 to control Wnt signaling transcriptional activity via regulation of TCF7L2 mRNA stability, forming a feedback loop that may contribute to cetuximab resistance and metastasis in CRC.

Triptolide decreases tumor-associated macrophages infiltration and M2 polarization to remodel colon cancer immune microenvironment via inhibiting tumor-derived CXCL12

Xuan Jiang et al.

Summary: Triptolide treatment shows therapeutic effects against colon cancer both in vivo and in vitro by inhibiting cancer cell proliferation and inducing apoptosis. In the tumor immune microenvironment, triptolide treatment reduces tumor-associated macrophage infiltration by downregulating tumor-derived CXCL12 expression, thus remodeling the immune microenvironment. Triptolide also inhibits the polarization of colon cancer cells to an anti-inflammatory state, and decreases the migration of cancer cells by reducing vascular endothelial growth factor expression.

HNRNPA2B1 regulates tamoxifen- and fulvestrant-sensitivity and hallmarks of endocrine resistance in breast cancer cells

Belinda J. Petri et al.

Summary: The RNA binding protein and m6A reader HNRNPA2B1 is overexpressed in breast tumors compared to normal tissue. Overexpression of A2B1 leads to tamoxifen and fulvestrant resistance, while knockdown of A2B1 restores endocrine sensitivity. Targeting A2B1 could be a potential therapeutic strategy to overcome acquired endocrine resistance in breast cancer.

Terpenoids' anti-cancer effects: focus on autophagy

Chirine El-Baba et al.

Summary: Terpenoids, the largest class of natural products derived mainly from plants, exhibit diverse anti-tumor effects, including anti-proliferative, apoptotic, anti-angiogenic, and anti-metastatic activities. The complex relationship between apoptosis and autophagy in cancer cells is driven by a delicate balance of protein regulation, leading to different outcomes through interrelated signaling pathways. Further research on autophagy induction by terpenoids may provide insights for novel cancer therapies and more effective treatment regimens.

Natural product triptolide induces GSDME-mediated pyroptosis in head and neck cancer through suppressing mitochondrial hexokinase-ΙΙ

Jing Cai et al.

Summary: The study reveals that triptolide eliminates head and neck cancer cells through inducing GSDME-mediated pyroptosis, and this effect is achieved by inhibiting HK-II and promoting caspase 3 activation. Combination with the SLC7A11 inhibitor erastin shows a strong synergistic effect in suppressing tumor survival in vitro and in a nude mice model. This research provides new insights into the use of TPL in cancer therapy.
