DpdtpA, A Multi-metal Ion Chelator, Attenuates Tau Phosphorylation and Microglial Inflammatory Response via Regulating the PI3K/AKT/GSK-36 Signal Pathways

Lu Wang et al.

Summary: Tau protein hyperphosphorylation and NFTs formation are key features of AD, and DpdtpA can attenuate these processes by regulating the PI3K/AKT/GSK-36 signaling pathway. DpdtpA also reduces microglial activation and inflammatory responses induced by Tau, offering a potential treatment option for AD-associated neuroinflammation.

Nfe2l2 Regulates Metabolic Rewiring and Epigenetic Reprogramming in Mediating Cancer Protective Effect by Fucoxanthin

Lujing Wang et al.

Summary: In this study, the role of Fucoxanthin (FX) and NFE2L2 in blocking tumor promoter TPA-induced skin cell transformation was evaluated using multi-omics approaches. The results showed that FX blocked TPA-induced oxidative stress and altered metabolisms, which were tightly coupled to epigenetic machinery. NFE2L2 played a critical role in FX-mediated metabolic rewiring and its potential consequences on epigenetic reprogramming. Overall, the findings suggest that the redox signaling of FX/NFE2L2 drives metabolic rewiring, causing epigenetic and transcriptomic reprogramming, potentially contributing to the protection against TPA-induced skin cancer.

Combination of paeoniflorin and calycosin-7-glucoside alleviates ischaemic stroke injury via the PI3K/AKT signalling pathway

Peng-Cheng Wang et al.

Summary: This study investigated the synergistic effects of PF + CG on ischaemia/reperfusion injury and found that PF + CG had significant protective effects in vivo and in vitro. It reduced neurobehavioral deficits, cerebral infarct volume, and brain edema, while increasing protective protein expression and suppressing apoptosis and oxidative stress. These findings suggest that PF + CG may be a potential treatment for ischaemic stroke.

Dysregulated phosphoinositide 3-kinase signaling in microglia: shaping chronic neuroinflammation

Erskine Chu et al.

Summary: Microglia play crucial roles in innate immunity within the mammalian central nervous system, but prolonged activation can lead to chronic neuroinflammation and neurodegenerative disease. Abnormalities in the PI3K-AKT signaling pathway in microglia may contribute to altered microglial activity and exacerbated neuroimmune responses. Modulation of microglial PI3K-AKT signaling shows promise as a therapeutic approach for various neuroinflammatory conditions.

The effect of aged microglia on synaptic impairment and its relevance in neurodegenerative diseases

Juan Jose Trivino et al.

Summary: Microglia play crucial roles in the CNS by regulating synapses and the neuronal network, responding to signals to protect the CNS, and influencing neuroinflammation and oxidative stress associated with aging and neurodegenerative diseases. Dysregulation of microglia function in aging may lead to cognitive dysfunction and increase susceptibility to neurodegeneration.

Rational Application of β-Hydroxybutyrate Attenuates Ischemic Stroke by Suppressing Oxidative Stress and Mitochondrial-Dependent Apoptosis via Activation of the Erk/CREB/eNOS Pathway

Yang Li et al.

Summary: Moderate dose of BHB enhances mitochondrial complex I activity, reduces oxidative stress, inhibits mitochondrial apoptosis, improves neurological function, and decreases infarct volume in stroke. These effects are achieved by upregulating SMCT1 and activating the Erk/CREB/eNOS pathway. This study provides a novel understanding of the neuroprotective effects of BHB in stroke.

AngIV-Analog Dihexa Rescues Cognitive Impairment and Recovers Memory in the APP/PS1 Mouse via the PI3K/AKT Signaling Pathway

Xiaojin Sun et al.

Summary: The study revealed that Dihexa can restore AngIV levels in the brain tissues of APP/PS1 mice by activating the PI3K/AKT signaling pathway, reducing inflammation, and increasing anti-inflammatory cytokine levels, thereby improving cognitive function and spatial learning. This suggests that the brain AngIV/PI3K/AKT axis may be a potential target for the treatment of AD.
