First discovered, long out of sight, finally visible: ribosomal DNA

Ashley N. Hall et al.

Summary: With the advancement of long-read sequencing, previously unresolvable genomic elements such as ribosomal DNA (rDNA) are being revisited. The variability in rDNA, including locus structure, copy number, and sequence variation, poses interesting questions about rDNA biology. Recent advances in long-read sequencing technology make it possible to incorporate the full rDNA sequence into reference genomes, allowing for a better understanding of rDNA biology within the context of cell physiology and whole-organism phenotypes.

Uneven Levels of 5S and 45S rDNA Site Number and Loci Variations across Wild Chrysanthemum Accessions

Jun He et al.

Summary: This study used oligonucleotide fluorescence in situ hybridization (Oligo-FISH) to draw ideograms of rDNA sites in Chrysanthemum accessions and found polymorphisms and co-localization of rDNA sites. There was also a correlation between the number of 5S rDNA sites and chromosome number, and the geographical distribution of different ploidy Chrysanthemum accessions may have promoted karyotype evolution. The study identified the formation mechanism of rDNA variations and provided a reliable method for examining rDNA variations in Chrysanthemum and related species, contributing to understanding the evolutionary and phylogenetic relationships of the Chrysanthemum genus.

Yuhuan Miao et al.

Summary: This study reported a complex chromosome-level genome assembly of Artemisia argyi, providing scientific support for evolutionary biology, functional genomics, and breeding. The assembly revealed whole-genome duplication events, transposable element bursts, and gene expansions related to photosynthesis, DNA replication, stress response, and secondary metabolism in A. argyi.

Thousands of high-quality sequencing samples fail to show meaningful correlation between 5S and 45S ribosomal DNA arrays in humans

Ashley N. Hall et al.

Summary: The study investigated copy number variation of ribosomal RNA genes (rDNA) and found no differences in probands with autism spectrum disorder and unaffected siblings. Validation results suggest that the previously reported concerted copy number variation between 45S and 5S arrays may be due to variable data quality, and meaningful correlation between 45S and 5S copy numbers was not detected in various datasets.
