Sophora alopecuroides Alleviates Neuroinflammation and Oxidative Damage of Parkinson's Disease In Vitro and In Vivo

Ting Sun et al.

Summary: Sophora alopecuroides L., a plant commonly used in northern China for food and herbal medicine, contains a new cytisine-type alkaloid called LY01, which has neuroprotective effects against Parkinson's disease (PD). PD is an irreversible neurodegenerative disease that seriously affects the elderly population. Current treatments have limited efficacy and numerous side effects, highlighting the need for new pharmacological options. This study evaluated the neuroprotective effects of LY01 and its underlying mechanisms using in vitro and in vivo models of PD.

PKCβII phosphorylates ACSL4 to amplify lipid peroxidation to induce ferroptosis

Hai-Liang Zhang et al.

Summary: Through CRISPR-Cas9 and kinase inhibitor screening, Zhang et al. found that PKC beta II phosphorylates and activates ACSL4 to enhance polyunsaturated fatty acid-containing lipid biosynthesis, thereby promoting accumulation of lipid peroxidation and ferroptosis.

Ferroptosis and its potential role in the physiopathology of Parkinson's Disease

Laura Mahoney-Sanchez et al.

Summary: Parkinson's Disease is a common neurodegenerative disorder with motor impairments and non-motor symptoms. Pathological features include dopaminergic neuronal death, alpha-synuclein deposition, iron accumulation, oxidative stress, and lipid peroxidation damage. Targeting pathways like ferroptosis could have therapeutic potential in treating PD.

Neuroprotective potential of cinnamon and its metabolites in Parkinson's disease: Mechanistic insights, limitations, and novel therapeutic opportunities

Efthalia Angelopoulou et al.

Summary: Cinnamon may have neuroprotective effects in neurodegenerative diseases, including Parkinson's disease. In vitro studies have shown that essential oils of cinnamon species can protect cells from oxidative stress, reactive oxygen species, and autophagy dysregulation, while in vivo studies have demonstrated that oral administration of cinnamon powder and sodium benzoate can protect against dopaminergic cell death and motor deficits.

Ferroptosis: mechanisms, biology and role in disease

Xuejun Jiang et al.

Summary: Ferroptosis, as a form of regulated cell death driven by iron-dependent phospholipid peroxidation, has seen significant growth in research in recent years. Studies have focused on molecular mechanisms, regulation, and functions of ferroptosis, linking this cell death modality to various pathologies and proposing its roles in normal physiology and potential therapeutic targeting.

PKC-β/Alox5 axis activation promotes Bcr-Abl-independent TKI-resistance in chronic myeloid leukemia

Dan Ma et al.

Summary: The study found that overexpression of PKC-beta is significantly correlated with TKI resistance in CML, with Alox5 and its related pathway playing a key role in this process by activating the ERK1/2 pathway. Inhibition of PKC-beta overexpression could potentially be a novel therapeutic mechanism to overcome TKI resistance in CML.
