Emerging Roles of Hedgehog Signaling in Cancer Immunity

Hedgehog signaling mechanism and role in cancer

Summary: Cell-cell communication through evolutionarily conserved signaling pathways plays a crucial role in embryonic development and adult tissue homeostasis. The Hedgehog (Hh) pathway, originally discovered in Drosophila, has been found to be involved in human development and diseases. Abnormal activation of the Hh pathway is implicated in various human cancers. Studies from model organisms and human patients provide insights into the Hh signal transduction mechanisms and offer potential strategies for targeting this pathway in cancer treatment.

Quinolines and Oxazino-quinoline Derivatives as Small Molecule GLI1 Inhibitors Identified by Virtual Screening

Summary: In this study, a virtual screening approach based on a five-feature pharmacophoric model was used to screen a database of commercially available compounds. The resulting quinoline derivatives showed significant reduction in GLI1 protein level and demonstrated submicromolar antiproliferative activity against human melanoma and medulloblastoma cell lines. Further structural modifications to the quinoline ring and chemical rigidification to an oxazino-quinoline scaffold provided insights for future ligand optimization.

Structure-based virtual screening identifies an 8-hydroxyquinoline as a small molecule GLI1 inhibitor

Summary: The GLI-family of transcription factors have emerged as a promising therapeutic target for human cancers, with GLI1 playing a central role in oncogenic signaling pathways. Compound 1, an 8-hydroxyquinoline, was identified as a high-affinity binder of GLI1 and found to inhibit GLI1-mediated transcriptional activity. Computational analyses suggested that compound 1 induces conformational changes in the GLI1 complex that prevent proper GLI function.
