Long-acting injectable PLGA/PLA depots for leuprolide acetate: successful translation from bench to clinic

Summary: Polyesters, with their excellent properties and ease of synthesis and modification, have been widely used in the pharmaceutical industry. The long-acting depots for leuprolide acetate, such as Lupron Depot (R) and ELIGARD (R), are among the first and most successful examples. Studying these products is of great importance for researchers in both academia and industry, as it will pave the way for the development of new and generic long-acting depots for various drugs.

Polymeric in situ forming depots for long-acting drug delivery systems

The use of biodegradable polymers like PLGA in ISFI systems allows for extended drug delivery through solvent exchange mechanisms, with drug release characterized by initial burst, diffusion processes, and polymeric degradation. Suitable solvent selection and consideration of PLGA hydrophobicity and molecular weights are crucial for optimizing drug release in ISFIs.
