RAB7A GTPase Is Involved in Mitophagosome Formation and Autophagosome-Lysosome Fusion in N2a Cells Treated with the Prion Protein Fragment 106-126

Summary: Failed communication between mitochondria and lysosomes can lead to dysfunctional mitochondria, potentially causing neurodegenerative diseases. This study demonstrates that RAB7A, a small GTPase, plays a crucial role in maintaining homeostasis in N2a cells treated with PrP106-126. Dysregulated RAB7A localization in mitochondria results in mitophagy deficiency, while overexpression of GTP-bound RAB7A promotes lysosome colocalization with mitochondria and protects mitochondrial homeostasis. These findings highlight the importance of RAB7A in the communication between mitochondria and lysosomes and suggest its potential as a target for modulating cellular homeostasis in neurodegenerative diseases.

Mitochondria ROS and mitophagy in acute kidney injury

Summary: Mitophagy is a crucial mitochondrial quality control mechanism that removes damaged mitochondria and reactive oxygen species (ROS) production. The relationship between mitochondrial oxidative stress, ROS production, and mitophagy is closely intertwined and involved in various pathological conditions of acute kidney injury (AKI). The complex process of mitophagy in mammals involves multiple pathways and its interaction with different types of cell death, such as apoptosis, pyroptosis, and ferroptosis in kidney injury, remains unclear. This review discusses recent advances in understanding the relationship between ROS and mitophagy, the different pathways of mitophagy, the interplay between mitophagy and cell death, and their significance in the pathogenesis of AKI.

Kidney injury molecule-1 and podocalyxin dysregulation in an arginine vasopressin induced rodent model of preeclampsia

Inhibiting Parkin expression significantly reduced mitophagy and increased mitochondrial injury and the rate of hepatocyte apoptosis under IH or IH with gAPN.
