Summary: In this study, the core promoters of five GST genes in Micromelalopha troglodyta were accurately identified and found to be significantly induced by two plant secondary metabolites in vitro. Two transcription factors, MtCncC and MtARNT, were cloned and shown to bind to the MtGSTt1 promoter and improve transcriptional activity, respectively. However, they had no regulatory function on the MtGSTz1 promoter.

Contribution of UDP-glycosyltransferases to chlorpyrifos resistance in Nilaparvata lugens

Summary: In this study, it was found that inhibiting the activity of UGT enzyme effectively increased the toxicity of chlorpyrifos to the chlorpyrifos-resistant strain of Nilaparvata lugens. The resistant strain showed higher content of UGT compared to the susceptible strain. Four UGT genes (UGT386H2, UGT386J2, UGT386N2, and UGT386P1) were significantly overexpressed in the resistant strain and could be induced by chlorpyrifos. Knockdown of UGT386H2 or UGT386P1 increased the susceptibility of the resistant strain to chlorpyrifos. These findings suggest that overexpression of these two UGT genes contributes to chlorpyrifos resistance in N. lugens.

Nuclear receptors potentially regulate phytochemical detoxification in Spodoptera litura

Summary: This study identified and analyzed 19 nuclear receptors (NRs) in the genome of Spodoptera litura larvae, and found that the transcriptional levels of most NRs were significantly induced in the midgut after exposure to flavone and xanthotoxin. Knockdown of four NRs (FTZ-F1, EcR, Dsf, and HR3) resulted in reduced larval tolerance to the phytochemicals. Additionally, many detoxification genes were downregulated by dsNR administrations, which may explain the high sensitivity of S. litura to phytochemicals. Molecular docking revealed that phytochemicals had high affinity to bind to NRs. This study provides insights into the mechanism of host adaptation in S. litura and supports the development of NR-targeted insecticides.

Glutathione S-Transferase Genes are Involved in Lambda-Cyhalothrin Resistance in Cydia pomonella via Sequestration

Summary: This study reports the involvement of GSTs in lambda-cyhalothrin resistance in the globally quarantined fruit pest Cydia pomonella. Most GST genes were upregulated in response to exposure to lambda-cyhalothrin, and their mRNA levels were considerably higher in a resistant population compared to susceptible strains. Recombinant CpGSTd3 was found to have the highest metabolic rate for lambda-cyhalothrin, supporting the role of GSTs in sequestration of the insecticide.
