Palladium(II)-Catalyzed Annulative Difunctionalization of Two Inert C(sp3)-H Bonds by a Bifunctional Reagent

Arghadip Ghosh et al.

Summary: This article presents a Pd-(II)-catalyzed direct difunctionalization of two C-(sp(3))-H bonds of aliphatic carboxylic acid derivatives using bifunctional reagents (BFRs) of the 2-iodobenzoic acid series. Various directing groups, ligands, bases, and co-oxidants are employed to facilitate the reaction, leading to the formation of benzo-fused peripheral-substituted caprolactones. Mechanistic studies provide insights into the crucial steps involved in the transformation. The versatility of the complex molecular entities is also showcased.

Chiral Arene Ligand as Stereocontroller for Asymmetric C-H Activation

Hao Liang et al.

Summary: This article reports the design and synthesis of a new type of readily tunable chiral arene ligands and their ruthenium(II) complexes, which have been successfully applied in enantioselective C-H activation. This study not only resolves the doubts about stereocontrol of chiral arene ligands in this reaction, but also opens up a new avenue for achieving asymmetric C-H activation.

Multiple annulations of inert C(sp2)-H bonds with alkynes

Arijit Saha et al.

Summary: Transition-metal catalyzed directing group assisted annulation of inert C-H bonds is a method for constructing complex molecular frameworks from readily accessible substrates. It allows the activation of directed inert C-H bonds and migration of transition metals, leading to the formation of structurally diverse fused pi-extended (hetero)cycles. The coordination preference of the directing group heteroatom determines the chemo- and regio-selectivity of the transformation. This study emphasizes the conceptual development of unsymmetrical multiple annulation of arene C(sp(2))-H bonds with alkynes, which is highly important.

Efficient Synthesis of Sulfur-Stereogenic Sulfoximines via Ru(II)-Catalyzed Enantioselective C-H Functionalization Enabled by Chiral Carboxylic Acid

Tao Zhou et al.

Summary: An undeveloped Ru(II)-catalyzed enantioselective C-H functionalization involving an enantiodetermining C-H cleavage step has been described in this study. Using a novel class of chiral binaphthyl monocarboxylic acids as chiral ligands, a broad range of sulfur-stereogenic sulfoximines were synthesized in high yields with excellent enantioselectivities (up to 99% yield and 99% ee) via desymmetrization, kinetic resolution, and parallel kinetic resolution. Furthermore, the resolution products can be easily transformed into chiral sulfoxides and key intermediates for kinase inhibitors.

Enantioselective Ruthenium-Catalyzed C-H Alkylations by a Chiral Carboxylic Acid with Attractive Dispersive Interactions

Uttam Dhawa et al.

Summary: The asymmetric ruthenium-catalyzed C-H alkylations were achieved using a chiral C2-symmetric carboxylic acid, resulting in chiral tetrahydrocarbazoles and cyclohepta[b]indoles with high enantioselectivity at room temperature. Mechanistic studies revealed a fast C-H ruthenation process, with a rate- and enantio-determining proto-demetalation step. The asymmetric induction was governed by weak attractive secondary dispersion interactions, as shown in NCI analysis of the key transition states.

Tatsuhiko Yoshino et al.

Summary: Enantioselective C-H functionalization is a powerful tool for synthesizing chiral molecules. The combination of high-valent group 9 metals with achiral Cp-x ligands and chiral carboxylic acids has shown promise in enabling selective cleavage and functionalization of enantiotopic C-H bonds. This Perspective provides a detailed summary of the background, catalyst design, applied reactions, and future directions in this field.
