Recent Progress in Chromium-Mediated Carbonyl Addition Reactions

Yuri Katayama et al.

Summary: Organochromium(III) species are versatile nucleophiles used in complex molecule synthesis due to their high tolerance of functional groups and chemoselectivity for aldehydes. Traditional methods of carbonyl addition reactions using organochromium(III) species require stoichiometric chromium(II) salts or catalytic chromium salts and stoichiometric reductants. Recent alternative methods involve organoradical formation from readily available starting materials and trapping the radical with stoichiometric or catalytic chromium(II) salts. These methods have the potential to develop sustainable chemical processes.

Merging Carbonyl Addition with Photocatalysis

Huan-Ming Huang et al.

Summary: The carbonyl group plays an important role in chemical reactions, and its combination with radicals through photoredox catalysis can lead to unique transformations. A novel strategy of using simple carbonyl compounds as intermolecular radical acceptors enables carbon-carbon and carbon-heteroatom disconnections. These reactions do not require metals, ligands, or additives, offering high atom economy. The combination of photoredox and metal catalysis has also been utilized to achieve notable transformations.

The Formal Cross-Coupling of Amines and Carboxylic Acids to Form sp3-sp3 Carbon-Carbon Bonds

Zirong Zhang et al.

Summary: A deaminative-decarboxylative protocol was developed to form new carbon(sp(3))-carbon(sp(3)) bonds using activated amines and carboxylic acids. Reductive nickel-catalyzed cross-coupling was utilized to leverage the building blocks, with miniaturized high-throughput experimentation studies critical for reaction optimization. The protocol is applied in late-stage diversification of pharmaceuticals, with over a thousand systematically captured and machine-readable reaction datapoints stored.

Direct Deamination of Primary Amines via Isodiazene Intermediates

Kathleen J. Berger et al.

Summary: The study reports a reaction that selectively deaminates primary amines and anilines under mild conditions with remarkable functional group tolerance, including pharmaceutical compounds, amino acids, amino sugars, and natural products. An anomeric amide reagent facilitates the reaction through the intermediacy of an unprecedented monosubstituted isodiazene intermediate, simplifying deamination and enabling strategic applications of iminium and amine-directed chemistries as traceless methods. Mechanistic and computational studies support the intermedicacy of a primary isodiazene, which exhibits unexpected divergence from previously studied secondary isodiazenes, leading to cage-escaping, free radical species undergoing a chain hydrogen-atom transfer process with aliphatic and diazenyl radical intermediates.

Carbonyl umpolung as an organometallic reagent surrogate

Xi-Jie Dai et al.

Summary: Construction of new carbon-carbon bonds is essential in organic chemistry, with organometallic reagents being versatile nucleophiles for this purpose. However, the classical organometallic reactions are limited by the dependence on stoichiometric quantities of metals and organic halides, leading to copious amounts of metal halide waste.

Reframing primary alkyl amines as aliphatic building blocks

Kathleen J. Berger et al.

Summary: While traditionally considered inert to substitution chemistry, primary aliphatic amines have been shown to be valuable C(sp(3)) synthons in recent advances in aliphatic deamination chemistry. Cross-coupling and photocatalyzed transformations using polar and radical mechanisms are compared with oxidative deamination and other transition metal catalyzed reactions.
