Kidney Damage Caused by Obesity and Its Feasible Treatment Drugs

Summary: The rapid growth of obesity worldwide has had a significant impact on the prevalence of chronic kidney disease, particularly in developing countries. Obesity has been strongly associated with chronic kidney disease and is an independent risk factor for kidney disease. Histological changes in obesity-induced renal injury include glomerular or tubular hypertrophy, focal segmental glomerulosclerosis or bulbous sclerosis. Inflammation, renal hemodynamic changes, insulin resistance, and lipid metabolism disorders are all involved in the development and progression of obesity-induced nephropathy. However, there is currently no targeted treatment for obesity-related kidney disease. This review presents the use of RAS inhibitors, SGLT2 inhibitors, and melatonin to treat obesity-induced kidney injury. Melatonin, in particular, has shown potential in protecting against kidney damage caused by obesity through inhibiting inflammation and oxidative stress.

Summary: This article investigates the role of the O-GlcNAc transferase (OGT)-STAT5 axis in regulating the immunomodulatory function and metabolic homeostasis of Treg cells. The results show that OGT deficiency in Treg cells makes mice more susceptible to high-fat diet-induced obesity and insulin resistance, while constitutive STAT5 activation in Treg cells protects against adipose tissue expansion and impaired glucose and insulin metabolism. Targeting the OGT-STAT5 axis can enhance Treg cell function to combat obesity and related metabolic disorders.

Specifically, individuals with obesity, particularly severe obesity, have a higher risk of poor outcomes from influenza and SARS-CoV-2 infections. T cells play a critical role in obesity-associated immune dysfunction, and altered T cell metabolism may contribute to this. Treatment with the metabolic drug metformin has shown promise in reversing abnormal T cell metabolism and improving outcomes to viral infection.

This highlights the important role of Treg-derived IL-10 in regulating adipose tissue homeostasis.

The pathogenic characteristics of these cells include increased chromatin accessibility, glycolytic output, and production of IL-17A, IFN-γ, and TNF-α. Modulation of glycolysis pathways can potentially reverse the inflammatory vigor of ihTh17 cells and severity of NAFLD.

RORC mRNA expression levels were found to be the best biomarker for discriminating non-allergic asthma among obese adolescents, hinting at a potential therapeutic target in obesity-related asthma. The NAO group exhibited higher TNFA mRNA expression levels as well as the highest IL17A and RORC mRNA expression levels among the different groups studied.

High-fat diet and dyslipidemia synergistically contribute to T cell senescence in gut associated lymphoid tissue

Summary: Recent studies have linked obesity with systemic T cell senescence and thymus involution, but the influence of high-fat diet (HFD) and diet composition on premature T cell senescence remains unclear. This study showed that HFD components synergistically induce T cell senescence in the gut-associated lymphoid tissue (GALT) with dyslipidemia. The results demonstrated a shift from naive to effector-memory (EM) phenotype in T cells and up-regulation of PD1 and KLRG1 in DIO mice, highlighting the complex interplay of HFD, obesity, and intestinal immunity.

However, these expressions showed no clear association with asthma control. Dexamethasone-induced CD4(+) FKBP51 expression was uniquely associated with worse asthma control in obese children with asthma, suggesting a potential mechanism for corticosteroid resistance in this population.
