Targeting the hallmarks of aging to slow aging and treat age-related disease: fact or fiction?

Maryam Keshavarz et al.

Summary: Aging is a major risk factor for various chronic diseases and finding ways to prevent or treat aging has become a research focus. The hallmarks of aging, including loss of proteostasis, mitochondrial dysfunction, altered nutrient sensing, and others, have been identified, but their role in shaping the aging rate is still debated. Future research should employ multidimensional analytical frameworks and appropriate study designs to further elucidate the interaction between these hallmarks and aging.

Estimating impact of food choices on life expectancy: A modeling study

Lars Fadnes et al.

Summary: This study developed a decision support model that predicts the impact of dietary choices on life expectancy. The optimal diet includes increased intake of whole grains, legumes, fish, fruits, and vegetables, while reducing red meat, processed meat, sugar-sweetened beverages, and refined grains. The transition from a typical Western diet to the optimized diet could significantly extend life expectancy.

Extending human healthspan and longevity: a symposium report

Loren M. DeVito et al.

Summary: Geroscience posits that aging is malleable and by targeting the hallmarks of biological aging, age-related diseases can be alleviated and longevity extended. Experts have made significant advancements in understanding the mechanisms underlying biological aging and identified ways to target aging pathways. Proof-of-concept studies and early-stage clinical trials are underway to assess the feasibility of drug evaluation in geroscience.

Long-term consumption of green tea EGCG enhances murine health span by mitigating multiple aspects of cellular senescence in mitotic and post-mitotic tissues, gut dysbiosis, and immunosenescence

Rohit Sharma et al.

Summary: Cellular senescence is a major hallmark of aging, and chronic consumption of green tea catechin epigallocatechin gallate (EGCG) can mitigate the progression of cellular senescence in adipose and intestinal tissues. EGCG attenuates DNA damage, inflammation, immunosenescence, and gut dysbiosis associated with aging, while promoting nutrient signaling mediators and preserving microbial diversity.

The Ground Zero of Organismal Life and Aging

Vadim N. Gladyshev

Summary: Cells can naturally or be forced to transition to a state of significantly lower biological age, or rejuvenation. These processes may converge to the mid-embryonic state, 'ground zero', where it is possible to achieve a biological age lower than naturally through genome editing and prolonging the rejuvenation phase.

Ursolic acid ameliorates stress and reactive oxygen species in C. elegans knockout mutants by the dopamine Dop1 and Dop3 receptors

Janine Nass et al.

Summary: Studies have shown that ursolic acid (UA) exerts neuroprotective effects through enhancing dopamine signaling, reducing the impact of stress-related disorders and extending lifespan. In vitro and in vivo experiments demonstrated strong antioxidant activity of UA, increased stress resistance in nematodes, and significant upregulation of dopamine receptor gene expression upon treatment with UA. The binding affinity of UA to dopamine receptors was stronger than dopamine itself, suggesting its potential as a lead compound for new antidepressant medications.

Autophagy Plays a Role in the Prolongation of the Life Span of Caenorhabditis elegans by Astaxanthin

Min Fu et al.

Summary: The study demonstrated that astaxanthin can prolong the lifespan of Caenorhabditis elegans, potentially through upregulating autophagy genes, which are regulated by both the insulin/IGF-1 signal pathway and TOR signal pathway.

Effect of citrus peels-supplemented diet on longevity, memory index, redox status, cholinergic and monoaminergic enzymes in Drosophila melanogaster model

Ganiyu Oboh et al.

Summary: The study demonstrated that citrus peels such as orange, tangerine, and grapefruit have potential life span promoting properties in Drosophila melanogaster. These citrus peels improved longevity, locomotor performance, memory, antioxidant status, and enzyme activities in treated flies. This suggests that citrus peels could be used as dietary supplements and functional food additives.

Caffeic and Dihydrocaffeic Acids Promote Longevity and Increase Stress Resistance in Caenorhabditis elegans by Modulating Expression of Stress-Related Genes

Sofia M. Gutierrez-Zetina et al.

Summary: Research shows that caffeic acid and dihydrocaffeic acid can increase the survival rate of Caenorhabditis elegans and extend the nematode's lifespan, potentially by activating multiple genes related to the insulin/IGF-1 pathway to produce beneficial effects on oxidative stress resistance and longevity.

Epigenetic clocks reveal a rejuvenation event during embryogenesis followed by aging

Csaba Kerepesi et al.

Summary: This study revealed a significant decrease in biological age, i.e., rejuvenation, during early stages of embryogenesis, followed by an increase in later stages. Pluripotent stem cells do not age, and the examined epigenetic age dynamics is conserved across species.

Longevity leap: mind the healthspan gap

Armin Garmany et al.

Summary: Life expectancy has increased by three decades since the mid-twentieth century, but healthspan expansion has not followed suit due to the prevalence of chronic diseases. Contemporary efforts are focused on extending health horizon by integrating public and societal endeavors with emerging interventions targeting age-related multimorbidity and frailty. This multidimensional approach, boosted by modern technologies, aims to bridge the healthspan-lifespan gap for a future of global wellbeing.
