Trypanosoma brucei RRP44: a versatile enzyme for processing structured and non-structured RNA substrates

Giovanna Cesaro et al.

Summary: Rrp44/Dis3 is a conserved eukaryotic ribonuclease that plays a role in processing and degradation of almost all types of RNA. Depletion of Rrp44 in Trypanosoma brucei blocks maturation of ribosomal RNA, leading to disruption of ribosome synthesis and inhibition of cell proliferation.

The interferon stimulated gene-encoded protein HELZ2 inhibits human LINE-1 retrotransposition and LINE-1 RNA-mediated type I interferon induction

Ahmad Luqman-Fatah et al.

Summary: Proteins encoded by interferon stimulated genes (ISGs) can inhibit LINE-1 retrotransposition. In this study, proteins that interact with the L1 ORF1-encoded protein (ORF1p) were identified using immunoprecipitation and mass spectrometry. Three ISG proteins were found to inhibit retrotransposition by interacting with ORF1p.

A glimpse on metazoan ZNFX1 helicases, ancient players of antiviral innate immunity

Giulia Blasi et al.

Summary: ZNFX1 is an interferon-stimulated protein found in the outer mitochondrial membrane, capable of binding dsRNAs and interacting with MAVS proteins to stimulate the early antiviral response. Phylogenetic analysis revealed a diverse distribution of ZNFX1 in metazoans, with structural conservation in the helicase region and presence of intrinsically disordered regions in selected proteins. Over-expression of ZNFX1 in bivalve and gastropod mollusks during dsDNA virus infection highlights its antiviral role, while functional diversification in other metazoan proteins remains to be determined.

Yeast Ssd1 is a non-enzymatic member of the RNase II family with an alternative RNA recognition site

Rosemary A. Bayne et al.

Summary: Ssd1, a conserved fungal RNA-binding protein, plays important roles in stress responses, cell division, and virulence. Through in vivo cross-linking and cDNA analysis, precise binding sites of Ssd1 on RNA were identified, particularly enriched in the 5' untranslated regions of mRNAs encoding cell wall proteins. The structure-guided mutations of Ssd1 prevent its binding to RNA, providing insights into the evolution of regulatory elements in the RNase II family.

Thousands of human non-AUG extended proteoforms lack evidence of evolutionary selection among mammals

Alla D. Fedorova et al.

Summary: By analyzing a large number of Ribo-seq datasets and genomic alignment data, this study reveals that the number of non-AUG initiated proteoforms greatly exceeds those with strong phylogenetic support. This suggests that the protein coding potential of the human genome is underestimated, and multiple proteins can be encoded by the same genomic loci.

Evolutionary and Expression Analysis of MOV10 and MOV10L1 Reveals Their Origin, Duplication and Divergence

Shuaiqi Yang et al.

Summary: This study found that MOV10 and MOV10L1 have some variations in zebrafish, but they may originate from a single ancestor. They showed different expression profiles in early development stages but similar patterns in adult zebrafish. Moreover, these genes were upregulated upon virus challenge, indicating their redundant conserved roles in virus infection.

Genome-wide bidirectional CRISPR screens identify mucins as host factors modulating SARS-CoV-2 infection

Scott B. Biering et al.

Summary: This study identified mucins as key host factors restricting viral infection, through genome-wide CRISPR knockout and activation screens in human lung epithelial cells expressing the SARS-CoV-2 entry factors ACE2 and TMPRSS2. These mucins serve as a significant viral restriction network inhibiting SARS-CoV-2 infection and other respiratory viruses. The findings provide a starting point for new host-directed therapeutics and highlight airway mucins as an important host defense mechanism.

Efficacy and safety of guanabenz acetate treatment for non-alcoholic fatty liver disease: a study protocol for a randomised investigator-initiated phase IIa study

Michihiro Iwaki et al.

Summary: Non-alcoholic fatty liver disease (NAFLD) is a liver disease associated with metabolic abnormalities. Currently, there is no consensus on the efficacy and safety of drugs used to treat NAFLD. Guanabenz acetate is a potential treatment for improving NAFLD and metabolic abnormalities. This study aims to evaluate the efficacy and safety of guanabenz acetate treatment in NAFLD patients.

Construction and Transcriptomic Study of Chicken IFNAR1-Knockout Cell Line Reveals the Essential Roles of Cell Growth- and Apoptosis-Related Pathways in Duck Tembusu Virus Infection

Chengwei Xiang et al.

Summary: This study reports the construction of a DF-1 cell line with knockout IFNAR1 gene, which enhances the replication of DTMUV and activates genes and signaling pathways related to cell growth and apoptosis. Knockdown of two up-regulated genes, HELZ2 and IFI6, also increases the replication of DTMUV RNA.

Impact of Zika virus on the human type I interferon osteoimmune response

Arnaud Drouin et al.

Summary: The field of osteoimmunology highlights the importance of the interferon (IFN) response pathway in osteoblasts. Research shows that Saos-2 cells are permissive to ZIKV and respond to IFN, with both viruses and IFN impacting the proteome of the cells in different ways. This study suggests that transient ZIKV infection can influence the osteoimmune state and that IFN and ZIKV activate distinctive proteomes in Saos-2 cells, potentially informing new therapeutic strategies.

Ribosome-Profiling Reveals Restricted Post Transcriptional Expression of Antiviral Cytokines and Transcription Factors during SARS-CoV-2 Infection

Marina R. Alexander et al.

Summary: The global COVID-19 pandemic has resulted in over 2.2 million deaths, highlighting the importance of understanding the host response to SARS-CoV-2. This study found that SARS-CoV-2 infection restricts the expression of various transcription factors and antiviral cytokine genes at the post-transcriptional level, potentially leading to high viral load and immune dysregulation.
