Computationally reconstructing cotranscriptional RNA folding from experimental data reveals rearrangement of non-native folding intermediates

Summary: The method called Reconstructing RNA Dynamics from Data (R2D2) is introduced to uncover details of cotranscriptional RNA folding. Through modeling the folding of Escherichia coli SRP RNA, it is found that specific local structural fluctuations are necessary for efficient cotranscriptional conformational rearrangement. Experiment-guided multi-scale modeling shows complex folding and processing of even a simple functional structure RNA during synthesis.
