Structural basis of Integrator-mediated transcription regulation

Isaac Fianu et al.

Summary: Integrator and PP2A collaboratively form a complex that dephosphorylates paused Pol II and cleaves nascent RNA to facilitate transcription termination. Integrator binds Pol II and pausing factors to exclude elongation factors, and positions PP2A to counteract Pol II phosphorylation. Additionally, Integrator endonuclease cleaves RNA near the Pol II active site, enabling DNA release and termination of transcription.

Structure of the human Mediator-RNA polymerase II pre-initiation complex

Srinivasan Rengachari et al.

Summary: Mediator is a conserved coactivator complex involved in the regulated initiation of transcription at eukaryotic genes. It interacts with transcriptional activators to stimulate RNA polymerase II phosphorylation and promoter escape. The complex structure involves various modules that can affect the activity conformation of CDK7 kinase.

Stochastic pausing at latent HIV-1 promoters generates transcriptional bursting

Katiana Tantale et al.

Summary: The study reveals that in latent HIV-1 cells, RNA polymerases undergo long-lived pausing, effectively limiting viral transcription. This pausing is stochastic rather than obligatory, with only a small fraction of polymerases experiencing long-lived pausing. The stochastic pausing leads to burst-like HIV-1 transcription in latent cells, facilitating the randomness of viral reactivation.

Quantitative imaging of transcription in living Drosophila embryos reveals the impact of core promoter motifs on promoter state dynamics

Virginia L. Pimmett et al.

Summary: TATA-containing and INR-containing promoters exhibit distinct dynamics in early Drosophila development, requiring one or two separate rate-limiting steps respectively, with TATA-driven promoters requiring more steps. Core promoter elements play an important role in regulating the rate limiting steps of transcription.
