The NFIB/CARM1 partnership is a driver in preclinical models of small cell lung cancer

Guozhen Gao et al.

Summary: CARM1 promotes transcription by modifying histones and chromatin bound proteins, and NFIB utilizes CARM1 as a coactivator. In small cell lung cancer, both CARM1 and methylated NFIB are critical for rapid tumor growth.

Bud31-mediated alternative splicing is required for spermatogonial stem cell self-renewal and differentiation

Junchao Qin et al.

Summary: This study reveals the crucial role of spliceosome component Bud31 in spermatogenesis in mice. Germ cell-specific knockout of Bud31 leads to male infertility. Bud31 is required for both spermatogonial stem cell pool maintenance and the initiation of spermatogenesis. The findings suggest that Bud31-mediated alternative splicing regulates the self-renewal and differentiation of male germ cells in mammals.

H3K36me2 methyltransferase NSD2 orchestrates epigenetic reprogramming during spermatogenesis

Zhiming Li et al.

Summary: This study reveals the important role of Nuclear receptor binding SET domain protein 2 (Nsd2) in spermatogenesis. The loss of Nsd2 leads to apoptosis and abnormal development of spermatogenic cells, as well as dysregulation of genes and histone modifications. Furthermore, Nsd2 deficiency results in the retention of histones and increased protamine proteins in spermatozoa. These findings uncover the involvement of Nsd2 in chromatin remodeling during spermatogenesis and provide insights into the molecular mechanisms of epigenetic abnormalities affecting male reproductive health.

SMYD5 catalyzes histone H3 lysine 36 trimethylation at promoters

Yanjun Zhang et al.

Summary: SETD2 methylates histone H3K36me3 in gene bodies in mammalian cells. In this study, the authors demonstrate that H3K36me3 is also enriched at promoter regions and is catalyzed by SMYD5, recruited by RNA polymerase II. They further show that SMYD5 is elevated in liver cancer, with a correlation to changes in gene expression. These findings provide insights into the localization and function of H3K36me3.

Splicing-associated chromatin signatures: a combinatorial and position-dependent role for histone marks in splicing definition

E. Agirre et al.

Summary: Through machine learning analysis, eleven chromatin modifications that differentially mark alternatively spliced exons have been identified. These marks act in a combinatorial and position-dependent way, forming characteristic splicing-associated chromatin signatures. The dynamic nature of chromatin modifications is proposed as a mechanism to rapidly fine-tune alternative splicing when necessary.

Sexual Dimorphism in Mouse Meiosis

Rong Hua et al.

Summary: Meiosis is a crucial process in gametogenesis of sexually reproducing organisms, but there are significant sex-specific differences within individual species in terms of chromatin reorganization, recombination, and tolerance for meiotic defects. Over the past two decades, numerous murine models have been developed to study the regulatory processes governing mammalian meiosis, particularly focusing on gene- and gametogenesis-related sexual dimorphism. This review aims to lay a solid foundation for future research on sex-specific differences in meiosis at the molecular level.

Chromatin-mediated alternative splicing regulates cocaine-reward behavior

Song-Jun Xu et al.

Summary: The study demonstrated the crucial role of H3K36me3 histone modification in alternative splicing, particularly in the context of mouse cocaine self-administration. Targeted epigenetic editing of H3K36me3 at Srsf11 directly influenced its alternative splicing and enhanced the effects of cocaine self-administration. These findings establish a direct causal relationship between H3K36me3, alternative splicing of Srsf11, and reward behavior.
