Interaction of the La-related protein Slf1 with colliding ribosomes maintains translation of oxidative-stress responsive mRNAs

Martin D. Jennings et al.

Summary: Cells respond to oxidative stress by reprogramming gene expression to enhance levels of antioxidant enzymes. In Saccharomyces cerevisiae, proteins Slf1 and Sro9 play a role in protein synthesis adaptation during stress. Slf1 is identified as a ribosome-associated translational modulator that promotes translation of highly-translated mRNAs, facilitating cell survival and adaptation to stress.

Cytosolic aspartate aminotransferase moonlights as a ribosome-binding modulator of Gcn2 activity during oxidative stress

Robert A. Crawford et al.

Summary: Translation regulation is an important cellular response to changing external conditions. This study identified RNA-binding proteins (RBPs) involved in translational control using polysome profiling and mass spectrometry. The results revealed previously under-appreciated RBPs and their roles in response to oxidative stress and amino acid starvation.

eIF4G is retained on ribosomes elongating and terminating on short upstream ORFs to control reinitiation in yeast

Mahabub Pasha Mohammad et al.

Summary: Translation reinitiation is a gene-specific translational control mechanism, where short upstream ORFs prevent full ribosomal recycling to allow the 40S subunit to resume traversing downstream. In yeast, eIF4G and eIF4A are preferentially retained on ribosomes elongating uORFs, and loss of eIF4G contact with eIF4A increases retention and efficiency of reinitiation on downstream initiation codons. Similar to humans, eIF4G is retained on ribosomes elongating uORFs to control reinitiation in yeast.
