Serotonergic circuit dysregulation underlying autism-related phenotypes in BTBR mouse model of autism

Molecular Abnormalities in BTBR Mice and Their Relevance to Schizophrenia and Autism Spectrum Disorders: An Overview of Transcriptomic and Proteomic Studies

Summary: Animal models play a crucial role in understanding the molecular basis of psychiatric disorders. The BTBR strain of mice, which exhibits characteristics of schizophrenia and autism spectrum disorders, has been extensively used as a model. However, the molecular mechanisms underlying the observed behavioral phenotypes in BTBR mice remain unclear. By analyzing transcriptomic and proteomic data from BTBR mice and comparing them with data from ASD and SCZ patients, we were able to identify brain-region-specific molecular abnormalities in BTBR mice and their relevance to human psychiatric disorders.

Summary: This study aimed to elucidate the neural circuits responsible for the regulation of social signaling response in BTBR mice, which showed reduced responsiveness to social stimuli and social signaling responses. The dysfunction of scent marking responses in BTBR mice was associated with blunted neuronal activity and morphological changes in selected brain areas, including the posterior bed nucleus of the stria terminalis and lateral habenula.

On the role of serotonin 5-HT1A receptor in autistic-like behavior: cross talk of 5-HT and BDNF systems

Summary: This study found that overexpression of the 5-HT1A receptor in the hippocampus of BTBR mice can reduce stereotyped behavior and increase time spent in the center in the open field test. However, it did not affect social behavior, immobility time, locomotor activity, and associative learning. Additionally, the overexpression of 5-HT1A receptor increased 5-HT7 receptor expression but decreased TrkB receptor expression.

Summary: This study using socially monogamous mandarin voles found that neural circuits involving 5-HTergic neurons in the dorsal raphe nucleus and serotonin terminals in the anterior cingulate cortex play a crucial role in consolation-like behaviors and sociability. The release of 5-HT in the ACC and activity of DR neurons were significantly increased during comforting behaviors and social interactions. Activation of 5-HT1A receptors in the ACC was able to reverse deficits in consolation and sociability induced by inhibition of 5-HTergic neurons in the DR.
