Modulation of GluA2-γ5 synaptic complex desensitization, polyamine block and antiepileptic perampanel inhibition by auxiliary subunit cornichon-2

Positive and negative allosteric modulation of GluK2 kainate receptors by BPAM344 and antiepileptic perampanel

Summary: In this study, cryo-EM structures of KAR GluK2 in complex with positive allosteric modulator BPAM344, competitive antagonist DNQX, and negative allosteric modulator perampanel were presented. The structures revealed the binding of two BPAM344 molecules per ligand-binding domain dimer interface. In the absence of an agonist or in the presence of DNQX, BPAM344 stabilized GluK2 in the closed state. The closed state was also stabilized by perampanel, which bound to the ion channel extracellular collar sites located in two out of four GluK2 subunits. These findings provide insights into the molecular mechanisms of positive and negative allosteric modulation of KARs.

Opening of glutamate receptor channel to subconductance levels

Summary: This study reveals the number of agonist binding domains required for ion channel opening and the impact of agonist binding on channel conductance in AMPA subtype ionotropic glutamate receptors. By combining structural analysis with experimental results, molecular dynamics simulations, and machine learning analysis, we found that at least two agonist binding domains are necessary for channel opening, and binding of agonist to all four domains does not guarantee maximal channel conductance.

Summary: In white matter, synaptic and axonal glutamatergic signaling to NG2 glia is critical for differentiation and myelination, whereas in gray matter the impact of neuron-to-NG2 glia signaling remains unclear. NG2 glia in hippocampus express AMPA receptors early in postnatal development, but the expression of TARP proteins changes during maturation. Adult NG2 glia receptors show increased sensitivity to blockers of Ca2+ permeable AMPA receptors, particularly near the soma.
