ColabFold: making protein folding accessible to all

Milot Mirdita et al.

Summary: ColabFold combines fast homology search and optimized model utilization to offer accelerated prediction of protein structures and complexes, with a processing speed that is 40-60 times faster. It serves as a free and accessible platform for protein folding, capable of predicting close to 1,000 structures per day.

Structural insights into GABA transport inhibition using an engineered neurotransmitter transporter

Deepthi Joseph et al.

Summary: This study investigates the structure and activity of GAT1 by modifying the dopamine transporter in Drosophila melanogaster. The findings suggest that the binding site undergoes substantial changes in order to accommodate GAT1 inhibitors, and an additional allosteric site is observed in the extracellular vestibule. The study also suggests a role for a specific motif in the selective control of neurotransmitter uptake.

Structural basis of GABA reuptake inhibition

Zenia Motiwala et al.

Summary: In this study, the structure of GAT1 protein in complex with the drug tiagabine was determined using cryo-electron microscopy. The results reveal that tiagabine inhibits GAT1 by blocking the intracellular gate and suppressing neurotransmitter uptake. This study provides important insights into the biology and pharmacology of neurotransmitter transporters and offers a blueprint for the design of neuromodulators.

Structural insights into the inhibition of glycine reuptake

Azadeh Shahsavar et al.

Summary: This study utilized synthetic single-domain antibodies and serial synchrotron crystallography to determine the structure of GlyT1 in complex with an inhibitor, revealing that the inhibitor locks GlyT1 in an inward-open conformation and binds at the intracellular gate of the release pathway. These findings define the mechanism of inhibition and provide insights for the rational design of new, clinically efficacious GlyT1 inhibitors.

Chloride-dependent conformational changes in the GlyT1 glycine transporter

Yuan-Wei Zhang et al.

Summary: The study found that chloride ions, along with sodium ions and glycine, stabilize an inward-open conformation of the human GlyT1 transporter, providing an unexpected insight into the role of chloride in this transporter family.

Illumination of serotonin transporter mechanism and role of the allosteric site

Dongxue Yang et al.

Summary: This study used cryo-electron microscopy structures to elucidate the structural states of SERT in different conditions, revealing all fundamental steps of the transport cycle. The research identified the substrate-bound allosteric site and provided new structural insights into how SERT controls the binding and unbinding of serotonin.

The antidepressant drug vilazodone is an allosteric inhibitor of the serotonin transporter

Per Plenge et al.

Summary: Vilazodone is an antidepressant with a unique binding pocket in the serotonin transporter (SERT) that exhibits non-competitive inhibition of serotonin uptake. This study combined pharmacology measurements and cryo-EM structural analysis to characterize Vilazodone binding to SERT, demonstrating its nanomolar affinity to an allosteric site in SERT.
