GABA signaling triggered by TMC-1/Tmc delays neuronal aging by inhibiting the PKC pathway in C. elegans

Summary: Aging is a major risk factor for neurodegenerative diseases, leading to functional decline and degeneration of neurons. Through a new method of neuronal proteomic profiling, certain proteins, including TMC-1, were found to have increased expression levels during aging, and TMC-1 was shown to have an anti-aging function in all tested neurons by regulating GABA signaling. Furthermore, the study demonstrated the vital role of metabotropic GABA receptors and G protein GOA-1 in the anti-neuronal aging functions of TMC-1 and GABA, and the activation of GABA receptors prevents neuronal aging by inhibiting the PLC ss-PKC pathway.

Structures of the TMC-1 complex illuminate mechanosensory transduction

Summary: This study reports the structure of the native TMC-1 mechanosensory transduction complex isolated from Caenorhabditis elegans. The complex is composed of TMC-1 subunits, CALM-1 calcium-binding protein, and TMIE transmembrane inner ear protein. CALM-1 makes extensive contacts with the TMC-1 subunits, while TMIE resides on the periphery of the complex.

Summary: The standard laboratory culture of Caenorhabditis elegans uses solid growth media with a bacterial food source, which limits control of food availability and worm population density. This study presents liquid-culture protocols for precisely modulating bacterial food availability and population density, allowing for reliable production of different types of worms. The protocols can be used for growing worms in small quantities for standard assays or in large quantities for other applications.
