Somatic mutations of CADM1 in aldosterone-producing adenomas and gap junction-dependent regulation of aldosterone production

Xilin Wu et al.

Summary: Somatic mutations of the neuronal cell adhesion gene CADM1 cause reversible hypertension by inhibiting gap junction communication and suppressing physiological aldosterone production. The intramembranous p.Val380Asp and p.Gly379Asp variants are key mutations associated with hyperaldosteronism and hypertension. This discovery has significant implications for the treatment of hypertension and primary aldosteronism.

ZnT1 is a neuronal Zn2+/Ca2+ exchanger

Noa Gottesman et al.

Summary: Research shows that ZnT1 regulates the transport of Zn2+ and Ca2+ in neurons, independent of pH gradients but dependent on Ca2+ gradients. ZnT1 serves as the primary efflux pathway for Zn2+ in neurons.

Summary: The HISTALDO consensus is useful to standardize nomenclature and achieve consistency among pathologists for the histopathologic diagnosis of unilateral PA. CYP11B2 immunohistochemistry should be incorporated into the routine clinical diagnostic workup to localize the likely source of aldosterone production.

Summary: This study demonstrates the application of targeted RNA sequencing to FFPE normal adrenal gland specimens, capturing distinct transcriptomic differences among adrenal zones. The data obtained will improve the understanding of transcriptomic patterns and potentially identify novel pathways controlling zone-specific steroid production.

Somatic mutations of GNA11 and GNAQ in CTNNB1-mutant aldosterone-producing adenomas presenting in puberty, pregnancy or menopause

Junhua Zhou et al.

Summary: The study identified gain-of-function somatic mutations in GNA11 or GNAQ in CTNNB1-mutant aldosterone-producing adenomas. These mutations were often found in patients during puberty, pregnancy, or menopause, with elevated LHCGR expression. While most aldosterone-producing adenomas have gain-of-function somatic mutations, the presence of these mutations in aldosterone-producing cell clusters of normal adrenal glands indicates a necessity for codriver mutations in APAs.
