Molecular Subtypes of Primary SCLC Tumors and Their Associations With Neuroendocrine and Therapeutic Markers

Song Qu et al.

Summary: This study validated a new molecular subtype classification for small cell lung cancer (SCLC) and identified intriguing associations between subtypes and therapeutic markers. It demonstrated significant intratumoral heterogeneity, with different subtypes of tumors exhibiting distinct immunophenotypes.

Genomic and transcriptomic analysis of a library of small cell lung cancer patient-derived xenografts

Rebecca Caeser et al.

Summary: The authors created patient derived xenograft models from 33 patients with small cell lung cancer and showed that these models retain the primary features and genomic landscapes of the original tumors through multi-omics sequencing. This resource of extensively annotated PDX/CDX models provides a powerful system to study the biology of small cell lung cancer.

Heterogeneity of neuroendocrine transcriptional states in metastatic small cell lung cancers and patient-derived models

Delphine Lissa et al.

Summary: This study explores the clinical significance and biological characteristics of neuroendocrine subtypes in small cell lung cancer, as well as the complex variability within and between tumors. NE tumors are more likely to respond to certain treatments but show resistance to chemotherapy, while non-NE tumors may benefit more from immunotherapy.

POU2F3 in SCLC: Clinicopathologic and Genomic Analysis With a Focus on Its Diagnostic Utility in Neuroendocrine-Low SCLC

Marina K. Baine et al.

Summary: POU2F3 is a recent marker of a small cell lung carcinoma (SCLC) subtype related to chemosensory tuft cells (SCLC-P). The expression of POU2F3 is associated with low expression of standard neuroendocrine markers and can be helpful in supporting the diagnosis of SCLC in cases with minimal or absent neuroendocrine marker expression. SCLC-P exhibits distinct genomic alterations that may provide a novel avenue for therapeutic targeting.

Patterns of transcription factor programs and immune pathway activation define four major subtypes of SCLC with distinct therapeutic vulnerabilities

Carl M. Gay et al.

Summary: Despite the heterogeneity of small cell lung cancer (SCLC), four subtypes have been identified based on tumor expression data, each showing different responses to treatment. SCLC-I subtype benefits the most from immunotherapy, while other subtypes have vulnerabilities to different inhibitors. Matching tumor subtype to therapy and manipulating subtype switching during treatment may enhance response depth and duration for SCLC patients.

Signatures of plasticity, metastasis, and immunosuppression in an atlas of human small cell lung cancer

Joseph M. Chan et al.

Summary: The study reveals a greater tumor diversity in SCLC compared to lung adenocarcinoma, with a PLCG2-high SCLC phenotype showing stem-like, pro-metastatic features and predicting worse overall survival. Furthermore, SCLC demonstrates greater immune sequestration and less immune infiltration than lung adenocarcinoma.

Small-cell lung cancer

Charles M. Rudin et al.

Summary: Small-cell lung cancer is a type of lung cancer characterized by high proliferative rate, early metastasis, and poor prognosis, strongly associated with tobacco carcinogens. Most patients are diagnosed with metastatic disease, with only a small fraction having potentially curative earlier-stage disease. Genomic profiling reveals extensive chromosomal rearrangements and high mutation burden in SCLC, often involving inactivation of TP53 and RB1 tumor suppressor genes.

Navin R. Mahadevan et al.

Summary: A subset of SCLC patients exhibit durable response to immune checkpoint blockade by upregulating MHC I after losing neuroendocrine differentiation, which can be enhanced through EZH2 inhibition and STING agonism, leading to immune rejection of SCLC in preclinical models.

Subtype heterogeneity and epigenetic convergence in neuroendocrine prostate cancer

Paloma Cejas et al.

Summary: Neuroendocrine carcinomas (NECs) arise from different anatomic sites, but share similar histological and clinical features. The epigenetic landscape of NECs converges towards a common state, while distinct subtypes occur within neuroendocrine prostate cancer, contributing to intratumor heterogeneity in clinical samples.

Summary: The study introduces a weighted-nearest neighbor analysis framework to learn the relative utility of each data type in each cell, enabling integrative analysis of multiple modalities. Applied to a CITE-seq dataset, the method constructs a multimodal reference atlas of the circulating immune system and successfully identifies and validates previously unreported lymphoid subpopulations.
