Distal meta-alkenylation of formal amines enabled by catalytic use of hydrogen-bonding anionic ligands

Summary: Regioselective and site-selective functionalization of distal C-H bonds has been a challenge due to the impact of covalently attached directing groups. Weak non-covalent interactions have been explored with Ir catalysis, but here we demonstrate a Pd-catalyzed meta-selective C-H functionalization of simple amines using anionic ligand and neutral substrate interactions. The site selectivity is governed by the H-bonding interaction between the anionic donating ligand and neutral motifs.

Palladium-Catalyzed Enantioselective β-C(sp3)-H Activation Reactions of Aliphatic Acids: A Retrosynthetic Surrogate for Enolate Alkylation and Conjugate Addition

Summary: CONSPECTUS: The development of asymmetric beta-C-H activation reactions for organic synthesis has been described in this Account. By studying reaction mechanisms and stereochemical properties, both strongly coordinating and weakly coordinating directing groups were designed to achieve beta-C-H activation of aliphatic acids and carboxylic acids, respectively. The development of five classes of chiral ligands enabled enantioselective beta-C-H activation reactions of readily available substrates.

Ligand-Enabled C-H Hydroxylation with Aqueous H2O2 at Room Temperature

This study introduces a new bifunctional bidentate ligand, CarboxPyridone, which enables the room-temperature Pd-catalyzed hydroxylation of benzoic and phenylacetic acids using aqueous hydrogen peroxide. The scalability and utility of this method is demonstrated through large-scale reactions and the synthesis of polyfluorinated natural products.

Synthesis of meta-substituted arene bioisosteres from [3.1.1]propellane

Summary: Small-ring cage hydrocarbons are popular bioisosteres for para-substituted benzene rings in drug design. In this study, we synthesized hydrocarbons called bicyclo[3.1.1]heptanes (BCHeps) from [3.1.1]propellane, which have geometric correspondence to meta-substituted benzenes. The BCHeps can be easily transformed into medicinally relevant carbon- and heteroatom-substituted compounds, displaying enhanced metabolic stability compared to their parent arene-containing drugs. This research demonstrates the potential of BCHeps as sp(3)-rich motifs in drug design and provides a new surrogate for meta-substituted benzene rings in drug discovery programs.

Advancing the Logic of Chemical Synthesis: C-H Activation as Strategic and Tactical Disconnections for C-C Bond Construction

Summary: The design of synthetic routes by retrosynthetic logic is influenced by available transformations. Transition-metal-catalyzed C-H activation is a powerful strategy for C-C bond formation, but its uptake in total synthesis has been tepid due to synthetic intractability and a lack of comprehensive guidelines. This Review addresses these issues and offers a guide to identify retrosynthetic opportunities to generate C-C bonds by C-H activation processes. Comparing traditional approaches and recent C-H activation methods, this Review demonstrates more efficient retrosynthetic strategies enabled by C-H activation for C-C bond construction.

Ligand-controlled divergent dehydrogenative reactions of carboxylic acids via C-H activation

The reactions allow chemoselective dehydrogenation of carboxylic acids, leading to the direct syntheses of alpha,beta-unsaturated carboxylic acids or gamma-alkylidene butenolides.

A tautomeric ligand enables directed C-H hydroxylation with molecular oxygen

Summary: This study reports a palladium complex that efficiently catalyzes hydroxylation of aryl carbon-hydrogen bonds at positions adjacent to carboxylic acids. The catalyst overturns the conventional site selectivity dictated by heterocycles, allowing late-stage modification of pharmaceutical compounds at previously inaccessible sites. Infrared, x-ray, and computational analysis support a potential role of ligand tautomerization in the catalytic cycle of this reaction.

The exploitation of transient chirality at sulfur was key to achieving stereochemical information transfer from ligand backbone to substrate carbons.
