Immunosuppressive microenvironment improvement and treatment of aggressive malignancy pancreatic ductal adenocarcinoma based on local administration of injectable hydrogel

Wenjia Zhang et al.

Summary: Pancreatic ductal adenocarcinoma (PDAC) is a devastating solid malignancy with a profuse fibrotic stromal reaction, hypoxia, and an immunosuppressive microenvironment. Immunotherapy has shown limited success in improving PDAC survival, so there is an urgent need for strategies that specifically target the immunosuppressive tumor microenvironment (TME) and immune-related therapeutic resistance. Photothermal immunotherapy, which combines local tumor hyperthermia with immune remodeling, has shown promise in PDAC treatment. This study develops an injectable peptide-based hydrogel integrated with near-infrared photothermal pigment and an immunomodulatory peptide for local PDAC administration and treatment, effectively inhibiting tumor recurrence and metastasis.

Mild photothermal therapy boosts nanomedicine antitumor efficacy by disrupting DNA mechanics damage repair pathways and modulating tumor

Yuxuan Xiong et al.

Summary: Various PARPi have been approved for the treatment of breast cancers, but they are only effective for HR-deficient breast cancers. The abnormal micro-environment of breast cancers hinders drug delivery. This study used mesoporous polydopamine to deliver PARPi and a DNA-damaging drug, and demonstrated that local mild photothermal therapy could inhibit the HR repair pathway and repress cancer-associated fibroblasts, thereby enhancing the antitumor effect and improving drug delivery.

Microbiotic nanomedicine for tumor-specific chemotherapy-synergized innate/adaptive antitumor immunity

Wencheng Wu et al.

Summary: This study reports a novel microbiotic nanomedicine LOD/TPZ@Lips-LA, which combines lactate oxidase (LOD) and prodrug tirapazamine (TPZ) loaded liposomes anchored on lactobacillus (LA) surface for chemotherapy-synergized antitumor immunity. The nanomedicine induces tumor cell apoptosis through in situ ROS generation and TPZ activation based on LA metabolism, resulting in strong immunogenic cell death (ICD). By regulating biomarker expression, it promotes anti-tumorigenic M1 polarization of innate tumor-associated macrophages and infiltration of adaptive cytotoxic CD8+ T cells, synergistically enhancing the tumor inhibition effects.

Improvement of radiotherapy with an ozone-carried liposome nano- system for synergizing cancer immune checkpoint blockade

Dan Zheng et al.

Summary: Radiotherapy synergizes with immune checkpoint blockade therapy by promoting immunogenic cell death. Researchers have developed an ozone-carrying liposome nano-system that can release ozone inside tumor cells, enhancing the anti-tumor efficacy of radiotherapy by inducing immunogenic cell death, releasing neoantigens, and relieving hypoxia symptoms.

Overcoming Challenges for CD3-Bispecific Antibody Therapy in Solid Tumors

Jim Middelburg et al.

Summary: CD3-bispecific antibody therapy is a rapidly developing field in immunotherapy, with approved use in hematological malignancies and ongoing research in solid tumors. However, treatment of solid tumors faces more challenges, including increased on-target off-tumor toxicities, sparse T-cell infiltration, and impaired T-cell quality due to an immunosuppressive tumor microenvironment, affecting the safety and efficacy of CD3-bispecific antibody therapy. Various combinatorial approaches are being explored to overcome these hurdles and improve the selectivity and effectiveness of the therapy.

PD-L1 as a biomarker of response to immune-checkpoint inhibitors

Deborah Blythe Doroshow et al.

Summary: PD-L1 expression is currently the best predictor of responsiveness to immune-checkpoint inhibitors, despite challenges in clinical use such as variability in different assays and cut-off points, as well as the lack of prospective comparisons with clinical outcomes. Nevertheless, several immunohistochemical assays have been approved for clinical use, with implications for pathology services and potential impacts on clinical outcomes. Comparison of FDA-approved PD-L1 assays, understanding of the varying implications of PD-L1 expression across different tumour types, and exploration of novel approaches to optimize its clinical utility are important topics for further research and practice.

Summary: MDSCs are pathologically activated cells with potent immunosuppressive activity, closely associated with poor clinical outcomes in diseases like cancer. Recent studies have identified key distinctions between MDSCs and classical neutrophils and monocytes, providing new insights into therapeutic targeting for cancer and autoimmune diseases through understanding their genomic and metabolic characteristics. Emerging data also suggest potential involvement of MDSCs in pregnancy, neonatal biology, and COVID-19.
