Ångstrom-resolution fluorescence microscopy

Susanne C. M. Reinhardt et al.

Summary: Fluorescence microscopy is a major characterization method in the life sciences, but current super-resolution techniques do not achieve Angstrom resolution. This study introduces a DNA-barcoding method that greatly improves the resolution of fluorescence microscopy, allowing for single-protein resolution in intact cells. The method also demonstrates Angstrom resolution for DNA origami.

Development of Deep-Learning-Based Single-Molecule Localization Image Analysis

Yoonsuk Hyun et al.

Summary: Recent developments in super-resolution fluorescence microscopic techniques (SRM) have led to nanoscale imaging, greatly enhancing our understanding of nanostructures. However, the performance of single-molecule localization microscopy (SMLM) is limited by the image analysis method. This review discusses the use of deep learning-based algorithms to improve SMLM image analysis and addresses future applications of deep learning methods for SMLM imaging.

Chromatin dynamics through mouse preimplantation development revealed by single molecule localisation microscopy

Marta Portela et al.

Summary: This study investigates the changes in chromatin during early development by using direct stochastic optical reconstruction microscopy (dSTORM) in mouse embryos. The researchers found spatial differences between cell types and their differentiation states. This analysis adds new information to previous biochemical studies and provides novel insights into chromatin organization during early development.

A role for nuclear stretching and NPCs changes in the cytoplasmic-nuclear trafficking of YAP: An experimental and numerical modelling approach

Stefania Saporito et al.

Summary: Mechanical forces play a significant role in eukaryotic cells, affecting cell shape, proliferation, polarity, and differentiation through mechanosensing and mechanotransduction. Recent findings suggest that cell-generated forces influence the translocation of transcription factors from the cytoplasm to the nucleus, which is influenced by the morphology of nuclear pore complexes. The complex patterns of nuclear envelope strains and stresses caused by cytoskeletal forces may alter the morphology of nuclear pore complexes, ultimately affecting molecular transport and cell functions.

Single molecule microscopy reveals key physical features of repair foci in living cells

Judith Mine-Hattab et al.

Summary: In response to double strand breaks, repair proteins accumulate at damaged sites, forming membrane-less sub-compartments or foci. Rad52 molecules freely diffuse within repair foci while Rfa1 molecules bind to ssDNA, suggesting the existence of a liquid droplet structure around damaged DNA.

Site-Specifically-Labeled Antibodies for Super-Resolution Microscopy Reveal In Situ Linkage Errors

Susanna M. Frueh et al.

Summary: In this study, a simple two-step protocol was developed to site-specifically attach reporters to IgG antibodies, and their performance in super-resolution microscopy techniques was evaluated. The results showed that errors from primary and secondary antibodies did not add up, and simulations revealed the main factors contributing to the errors.

Three-dimensional superresolution fluorescence microscopy maps the variable molecular architecture of the nuclear pore complex

Vilma Jimenez Sabinina et al.

Summary: NPCs are large macromolecular machines that regulate traffic between the nucleus and the cytoplasm, consisting of about 1000 proteins and having a mass of over 100 MDa in vertebrates. While a static model of the central scaffold of the NPC has been assembled, many structural components remain unknown due to the NPC's size and flexibility. This study combined 3D super-resolution microscopy with computational classification to create a detailed 3D structural map of the NPC in single human cells, providing insight into the different conformations of the nuclear ring and the nuclear basket.
