Genomic mutation landscape of skin cancers from DNA repair-deficient xeroderma pigmentosum patients

Andrey A. Yurchenko et al.

Summary: Xeroderma pigmentosum (XP) is a rare genetic disorder associated with an increased risk of skin cancer. In this study, the authors analyze skin cancer genomes from five different XP groups, revealing genetic and molecular factors related to the mutational profile and UV-induced mutagenesis in XP.

Spontaneous mutagenesis in human cells is controlled by REV1-Polymerase z and PRIMPOL

Zsolt Gyure et al.

Summary: Translesion DNA synthesis (TLS) using specialized DNA polymerases facilitates replication over damaged or difficult-to-replicate templates. Investigating three main TLS control mechanisms, REV1 and PCNA ubiquitylation recruit TLS polymerases while PRIMPOL creates post-replicative gaps. Whole-genome sequencing of human RPE-1 cell clones reveals REV1 and Polymerase z as responsible for base substitution mutagenesis resembling homologous recombination deficiency, while PRIMPOL-/- cells show reduced component resembling oxidative mutagenesis. Small deletions in short repeats appear in REV1-/-PCNAK164R/K164R double mutants, revealing an alternative TLS mechanism. REV1-/- and REV3L-/- mutants exhibit 500-5,000 bp deletions, and REV1-/- PRIMPOL-/- cells show chromosomal instability. TLS protects against deletions and large rearrangements but contributes to spontaneous base substitutions.

DNA Damage Tolerance Pathways in Human Cells: A Potential Therapeutic Target

Ashlynn Ai Li Ler et al.

Summary: DNA damage tolerance pathways play a crucial role in allowing replication to proceed in the presence of lesions. Bypassing DNA lesions by TLS polymerases can lead to mutagenesis and cancer development, while homology-directed DDT allows error-free lesion bypass. Understanding these pathways can provide therapeutic avenues.

DNA polymerase eta protects human cells against DNA damage induced by the tumor chemotherapeutic temozolomide

Marcela T. Latancia et al.

Summary: This study investigates the effects of TMZ treatment on human XP-V cells and its complemented counterpart. The results show that pol eta plays a role in protecting human cells from TMZ-induced DNA damage, but this role may be different from its canonical TLS mechanism. This finding opens up new therapeutic possibilities of using pol eta as a target to improve the efficacy of TMZ-based cancer therapies.

Changes in the architecture and abundance of replication intermediates delineate the chronology of DNA damage tolerance pathways at UV-stalled replication forks in human cells

Yann Benureau et al.

Summary: Research has found that DNA lesions in S phase can threaten genome stability, but specific damage tolerance mechanisms can overcome these obstacles and ensure the continuity of DNA synthesis. Specifically, the TLS polymerase eta plays a crucial role at the replication fork, and RAD51 also supports repriming at the replication fork, providing important clues for addressing issues caused by DNA damage.

Translesion synthesis inhibitors as a new class of cancer chemotherapeutics

Seema M. Patel et al.

Summary: Translesion synthesis (TLS) is a DNA damage tolerance mechanism that plays a primary role in replicating past DNA lesions induced by genotoxic agents in cancer. Inhibitors targeting various TLS DNA polymerases or key protein-protein interactions in the TLS machinery have shown promise as a new class of anti-cancer adjuvant agents. Further studies are needed to determine the clinical potential and optimal targets for TLS inhibitors in combination therapies with current genotoxic chemotherapies.

The major mechanism of melanoma mutations is based on deamination of cytosine in pyrimidine dimers as determined by circle damage sequencing

Seung-Gi Jin et al.

Summary: The study identified cytosine-deaminated CPD as the primary premutagenic lesion responsible for mutations in melanomas. These lesions were enriched immediately upstream of transcription start sites, indicating a potential mutation-promoting role of bound transcription factors. The genomic sequence specificity of deaminated CPDs matched the mutation signature of melanomas.
