The Fundamental Role of Lipids in Polymeric Nanoparticles: Dermal Delivery and Anti-Inflammatory Activity of Cannabidiol

Summary: This study presents a nanoparticulate platform for the topical treatment of inflammatory conditions using cannabidiol (CBD). Lipid-stabilized nanoparticles (LSNs) were shown to effectively encapsulate CBD and suppress the release of IL-6 and IL-8. Different stabilizing lipids, including cetyl alcohol, stearic acid, lauric acid, and a combination of stearic acid and lauric acid, were examined for their effects on CBD release and skin permeation.

Effects of splenectomy on skin inflammation and psoriasis-like phenotype of imiquimod-treated mice

Summary: This study analyzed the cellular composition of spleen and measured metabolites in blood of IMQ-treated mice, as well as investigated the influence of splenectomy on the degree of skin inflammation and pathology. The results showed that the composition of cells in the spleen and blood metabolites positively correlated with the weight of the spleen. However, splenectomy did not affect the psoriasis-like phenotypes in IMQ-treated mice.

Application of imiquimod-induced murine psoriasis model in evaluating interleukin-17A antagonist

Summary: Our study developed a 7-day psoriasis model in female C57BL/6 mice with optimized usage of IMQ, positive control reagent, and anti-mIL17A antibody, providing a reliable tool for evaluating IL17A antagonists. We demonstrated the involvement of IL17A signaling in disease progression, showing that IMQ induced the expression of IL17A and related cytokines, and their downstream cytokines could be inhibited by IL17A antagonists. Additionally, we found that IMQ transiently induced IL17A, directly leading to downstream signaling activation, highlighting the importance of understanding the kinetic changes in IL17A signaling for the investigation of IL17A antagonists.

Cannabidiol selectively modulates interleukin (IL)-1β and IL-6 production in toll-like receptor activated human peripheral blood monocytes

Summary: This study evaluated the impact of CBD on human primary monocytes activated through Toll-like receptors (TLR). The results showed that CBD suppressed the secretion of inflammatory cytokine IL-1β by monocytes activated through most TLRs, while also significantly modulating the production of IL-6. Most other immune mediators were not affected by CBD modulation.

Characterization of nanoparticles made of ethyl cellulose and stabilizing lipids: Mode of manufacturing, size modulation, and study of their effect on keratinocytes

Topical gel based nanoparticles for the controlled release of oleanolic acid: design and in vivo characterization of a cubic liquid crystalline anti-inflammatory drug

Summary: Oleanolic acid (OA) has anti-inflammatory properties but limited permeability, so a cubic liquid crystalline nanoparticle (LCNP) gel was prepared as a topical delivery system for OA. The optimized LCNP formulation showed superior drug release and permeation compared to a standard gel, with controlled release mechanisms identified through studies. In a rodent inflammation model, the OA-LCNP gel demonstrated sustained efficacy after a single application.
