Targeted therapy for lung cancer: Beyond EGFR and ALK

Mercedes Herrera-Juarez et al.

Summary: Precision oncology aims to design the best cancer treatment based on tumor biology. In NSCLC, patients with actionable genomic aberrations can benefit from targeted therapy, such as EGFR mutations and ALK rearrangements. Effective inhibitors have been developed for various druggable targets, leading to a paradigm shift in NSCLC treatment.

Early in vitro evidence indicates that deacetylated sialic acids modulate multi-drug resistance in colon and lung cancers via breast cancer resistance protein

Isaac Tuffour et al.

Summary: Cancers utilize sugar residues to engage in multidrug resistance, specifically involving the glycan sialic acid (Sia) and its functional group alterations. This study investigated the role of deacetylated-Sias in the modulation of multidrug resistant pathways in colon and lung cancer cells via the BCRP transporter protein. The findings suggest that deacetylated Sia is utilized by colon and lung cancers to engage in MDR through overexpression and efflux action of BCRP.

Antiproliferation and apoptosis studies of estrone pharmacophores in triple-negative breast cancer

Felix Acheampong et al.

Summary: In this study, it was discovered that estrone analogs with novel pharmacophores have high potency against triple-negative breast cancer (TNBC) cells. These analogs inhibit cell proliferation by targeting the EGFR receptor and induce mitochondrial apoptosis. This research provides a foundation for further development of estrone derivatives as novel therapeutics for TNBC.

Triazole-estradiol analogs: A potential cancer therapeutic targeting ovarian and colorectal cancer

Trevor Ostlund et al.

Summary: 1,2,3-triazoles and their combination with steroid skeletons as drug delivery systems have potential therapeutic effects against various cancers. In vitro studies on ovarian and colorectal cancer cells demonstrated their inhibitory activities and their ability to inhibit proteins in the EGFR pathway. They also showed inhibition against drug efflux proteins associated with drug resistance.

Randomised controlled trial of first-line tyrosine-kinase inhibitor (TKI) versus intercalated TKI with chemotherapy for EGFR-mutated nonsmall cell lung cancer

Rolof G. P. Gijtenbeek et al.

Summary: This study found that in patients with EGFR-mutated NSCLC, intercalated erlotinib with cisplatin-pemetrexed chemotherapy significantly prolonged progression-free survival compared to erlotinib monotherapy, but at the cost of increased toxicity.

Regulation of Cell Cycle Progression by Growth Factor-Induced Cell Signaling

Zhixiang Wang

Summary: The cell cycle is a series of events driving cell division and production of two new daughter cells, involving phases G1, S, G2, and M. Progression is mediated by Cdks and their cyclin subunits, but growth factor signaling pathways are the driving force. Role of growth factor signaling in phases other than G1 is still poorly understood, with research focusing on regulation of cell cycle progression through different phases.

Deacetylated sialic acids modulates immune mediated cytotoxicity via the sialic acid-Siglec pathway

Susan Grabenstein et al.

Summary: Cancers exploit glycans through the Sia-Siglec pathway to evade the immune system, with atypical forms of sialic acid binding to Siglec receptors on NK cells. Modulating acetylation levels of Neu5Ac through SIAE and CASD1 genes influences Siglec binding in various cell lines, affecting NK cell mediated cytotoxicity and potential cancer immune therapies.
