Future Antimicrobials: Natural and Functionalized Phenolics

Summary: With the increasing incidence of global antimicrobial resistance, the development of new antimicrobial molecules is needed. Phenolic compounds have a versatile scaffold that allows for chemical additions and exhibit potent antimicrobial activities that can be enhanced through functionalization. Functionalization through esterification, phosphorylation, hydroxylation or enzymatic conjugation can increase the antimicrobial activity of phenolic compounds and reduce the required minimal concentrations. Phenolics target multiple sites in bacteria, interfering with cell wall synthesis, DNA replication, or enzyme production, leading to higher sensitivity of bacterial cells towards these natural compounds. This review summarizes the important knowledge on functionalization of natural phenolic compounds and their effects on antimicrobial activity.
