Amyrel, a novel glucose-forming α-amylase from Drosophila with 4-α-glucanotransferase activity by disproportionation and hydrolysis of maltooligosaccharides

Summary: The alpha-amylase paralogue Amyrel found in true flies is unique among animals as it possess both hydrolytic alpha-amylase activity and a 4-alpha-glucanotransferase transglycosylation activity. Amyrel specifically reacts on alpha-(1-4) glycosidic bonds of starch, producing a complex mixture of maltooligosaccharides. The main structural determinant of these atypical activities is a Gly-His-Gly-Ala deletion in the flexible loop bordering the active site, providing more freedom to the substrate for subsites occupancy and allowing a less-constrained action pattern resulting in versatile activities at the active site.
