The Colorimetric Detection of the Hydroxyl Radical

Summary: Investigation was made on an aromatic substrate for hydroxylation by hydroxyl radicals ((OH)-O-center dot). The probe, N,N'-(5-nitro-1,3-phenylene)-bis-glutaramide, and its hydroxylated product were found not to bind either iron(III) or iron(II), thus not interfering with the Fenton reaction. A spectrophotometric assay based on the hydroxylation of the substrate was developed, providing univocal and sensitive (OH)-O-center dot detection. The assay demonstrated the lack of Fenton activity of iron(III) complexes of long-chain fatty acids under biological conditions.

Highly Selective Fluorescent Probe With an Ideal pH Profile for the Rapid and Unambiguous Determination of Subcellular Labile Iron (III) Pools in Human Cells

Recent Advances in the Elucidation of Frataxin Biochemical Function Open Novel Perspectives for the Treatment of Friedreich's Ataxia

Summary: Friedreich's ataxia (FRDA) is a prevalent autosomic recessive ataxia that is associated with severe cardiac hypertrophy and less commonly, diabetes. It is caused by mutations in the gene encoding frataxin, a mitochondrial protein involved in iron metabolism. Recent discoveries suggest that frataxin functions as an accelerator in the biosynthesis of Fe-S clusters, which may have implications for the development of therapeutic treatments for FRDA.

A new fluorescent sensor mitoferrofluor indicates the presence of chelatable iron in polarized and depolarized mitochondria

Summary: We synthesized a new fluorescent indicator, MFF, to measure mitochondrial chelatable iron in living cells. MFF accumulates in polarized mitochondria and remains even after depolarization. MFF fluorescence colocalizes with the mitochondrial membrane potential indicator and can be affected by iron delivery agents and metal chelators.

Summary: The study shows that glutathione is not the cellular ligand for heme and its presence can enhance heme degradation. The concentration of heme in the cytosol is extremely low and intracellular heme transfer may occur via intracellular membrane structures. If any heme escapes into the cytosol, it will quickly bind to glutathione for protection against its toxic effects.
