Cosolvent Dimethyl Sulfoxide Influences Protein-Ligand Binding Kinetics via Solvent Viscosity Effects: Revealing the Success Rate of Complex Formation Following Diffusive Protein-Ligand Encounter

Sven Wernersson et al.

Summary: Protein-ligand-exchange kinetics is crucial for the duration of biochemical signals and its role in drug design. This study investigates the effect of the solvent dimethyl sulfoxide (DMSO) on protein-ligand binding. Results show that increasing DMSO concentration leads to decreased affinity and slower association rate, while dissociation rate is less affected. Only a small fraction of binding attempts result in successful complex formation.

Solubility of amlodipine besylate in binary mixtures of polyethylene glycol 400+water at various temperatures: Measurement and modelling

Elmira Behboudi et al.

Summary: Measuring and evaluating the solubility of amlodipine besylate (AMB) in a binary solvent system of water and polyethylene glycol 400 (PEG 400) was carried out in this study. The solubility of AMB increased nonlinearly with increasing the mass fraction of PEG 400. The best correlation results were obtained using the CNIBS/R-K and NRTL models. The dissolution and mixing thermodynamic properties of AMB in the aqueous PEG 400 mixtures were also calculated and the density values of the mixtures were measured.

Crystal Engineering of Pharmaceutical Cocrystals in the Discovery and Development of Improved Drugs

Geetha Bolla et al.

Summary: Crystal engineering is a discipline that started in the 1970s, focusing on the study of topochemical reactions in the solid state. One important application of crystal engineering is in the design of pharmaceutical cocrystals, which can systematically tune the physicochemical properties of drugs without altering their molecular structure. This review traces the development of supramolecular synthons as robust hydrogen bond patterns for pharmaceutical cocrystal design, and discusses recent advancements in the field that have led to the launch of several drugs with improved efficacy and bioavailability. The importance of pharmaceutical cocrystals lies in their ability to bridge the gap between drug discovery and development, and their potential to provide high-value pharmaceuticals with economic and environmental benefits.

Hirshfeld surface and electrostatic potential surface analysis of clozapine and its solubility and molecular interactions in aqueous blends

Xinding Yao et al.

Summary: This paper focuses on the solubility, thermodynamic properties, and molecular interactions of clozapine in aqueous solutions with different protic and aprotic cosolvents. The solubility variation of clozapine is mainly controlled by hydrogen-bonding basicity, solubility parameter, and dipolarity-polarizability of the solvents. Various quantitative analysis methods were used to correlate the solubility data and study the solvation behavior. Furthermore, the preferential solvation of clozapine by solvent species in solutions and the thermodynamic properties of dissolution were investigated and discussed.

Mapping the energy landscape of protein-ligand binding via linear free energy relationships determined by protein NMR relaxation dispersion

Olof Stenstrom et al.

Summary: Biochemical signaling is mediated by complexes between macromolecular receptors and their ligands, with drug efficacy in vivo depending on the lifetime of the drug-protein complex. The molecular details of designed ligands affect binding kinetics and equilibrium binding constant. Ligands with reduced off-rates primarily stabilize the bound state, with the transition state located closer to the free state, suggesting a critical role of desolvation. Different subclasses of ligands show different behavior in this regard.
