Aryl-n-hexanamide linked enaminones of usnic acid as promising antimicrobial agents

Summary: Lichen secondary metabolite usnic acid is a potential antitubercular and antibacterial molecule. Through designing, synthesizing, and evaluating a set of usnic acid derivatives, several compounds with potential antitubercular and antibacterial activity were discovered. These pharmacological results will contribute to the development of usnic acid derivatives as potential antimicrobial agents.

Docking and scoring for nucleic acid-ligand interactions: Principles and current status

Summary: Nucleic acid-ligand interactions play crucial roles in cellular processes and have attracted significant interest in drug discovery. However, traditional docking algorithms and scoring functions for protein-ligand interactions may not be applicable to nucleic acid-ligand docking due to the differences in their properties. This review discusses the current status, challenges, and limitations of docking algorithms and scoring functions for DNA/RNA-ligand interactions.

Cascade synthesis of indolizines and pyrrolo[1,2-a]pyrazines from 2-formyl-1-propargylpyrroles

Summary: The straightforward synthesis of indolizines and pyrrolo[1,2-a]pyrazines was achieved through a cascade condensation/cyclization/aromatization reaction. The reaction proceeded satisfactorily under basic conditions, providing high yields of the desired products. N-Allenyl-2-formylpyrroles were found to be more reactive substrates compared to 2-formyl-N-propargylpyrroles in this reaction system.

Antifungal Activity of Fibrate-Based Compounds and Substituted Pyrroles That Inhibit the Enzyme 3-Hydroxy-methyl-glutaryl-CoA Reductase of Candida glabrata (CgHMGR), Thus Decreasing Yeast Viability and Ergosterol Synthesis

Summary: Due to the emergence of multidrug-resistant strains of yeasts, discovering antifungal agents that target alternative molecular targets is urgently needed. This study evaluated the capacity of three different series of synthetic compounds to inhibit a specific enzyme in Candida glabrata, leading to the disruption of ergosterol synthesis and yeast viability. Two compounds, 1c and 5b, were identified as the best antifungal candidates among the compounds studied. They effectively inhibited the growth of fluconazole-resistant and fluconazole-susceptible C. glabrata strains. The compounds act by inhibiting the mevalonate synthesis pathway, resulting in a decrease in ergosterol concentration. The docking analysis suggested that their inhibitory effect may be mediated by their interaction with the enzyme's catalytic site. Further research and modifications to increase their specificity and potency are needed.

This review focuses on gold-catalyzed reactions of enaminones and recent advances, along with their interesting mechanistic aspects.
