Quercetin prevents osteoarthritis progression possibly via regulation of local and systemic inflammatory cascades

Summary: Due to a lack of effective treatments, osteoarthritis (OA) remains a challenge for clinicians. Quercetin, a bioflavonoid, has shown potent anti-inflammatory effects, but its effect on preventing OA progression and the underlying mechanisms are still unclear. In this study, rats were divided into different groups and treated with quercetin for 28 days. The results showed that high-dose quercetin had a protective effect against OA development and progression by inhibiting the expression of inflammatory factors and promoting the expression of cartilage-related proteins. Quercetin could be a potential therapeutic agent for preventing OA progression in at-risk groups.

Development of flavonoid probes and the binding mode of the target protein and quercetin derivatives

The results suggest that quercetin and 3-O-methylquercetin can inhibit the activity of matrix metalloproteinase-1 (MMP-1), and their interaction with the catalytic domain of MMP-1 is dose-dependent. Analysis using 15N-labeled MMP-1 catalytic domain reveals that 3-O-methylquercetin interacts with metal ions in MMP-1. The development of flavonoid probes can help discover new target proteins and elucidate the core mechanisms of flavonoids' multiple bioactivities.

Further clinical studies considering different confounding factors are needed to provide evidence for the actual impact of dietary flavonoids on human diseases. One possible mechanism involves the regulation of reactive oxygen species and 5'-adenosine monophosphate-activated protein kinase by flavonoids.
