Optimization of the supercritical extraction of rosmarinic acid from clary sage residue and the antioxidant activity of the extracts

Summary: This study investigates the extraction of bioactive compounds from clary sage distillation residue using carbon dioxide under supercritical conditions. The target compound, rosmarinic acid, is a naturally occurring phenolic compound with strong antioxidant properties. Response surface methodology is employed to optimize the extraction parameters, including temperature, pressure, and co-solvent composition. The results demonstrate that the extraction yield and antioxidant activity of the extracts vary significantly under different extraction conditions. Based on the optimization study, the optimal conditions are determined to be 100 bar, 65 degrees C, and 35% ethanol co-solvent, resulting in a rosmarinic acid concentration of more than 8 mg/g.

Ultrasound-assisted extraction and properties of polysaccharide from Ginkgo biloba leaves

Summary: Response surface methodology was used to optimize the extraction conditions of Ginkgo biloba leaves polysaccharide (GBLP) assisted by ultrasound. The optimum conditions were determined as 30 mL/g liquid to material ratio, 340 W ultrasonic power, and 50 min extraction time, resulting in a GBLP yield of 5.37%. Two chemically modified polysaccharides, CM-GBLP and Ac-GBLP, were obtained from GBLP and their physicochemical properties and in vitro antioxidant activities were compared. The results showed improved solubility and antioxidant activity for the chemically modified polysaccharides, indicating the effectiveness of carboxymethylation and acetylation methods in enhancing polysaccharide properties.

Coordinating caffeic acid and salvianic acid A pathways for efficient production of rosmarinic acid in Escherichia coli

Summary: A new biosynthetic pathway was constructed by introducing caffeic acid synthesis modules into Escherichia coli, leading to an increased titer of rosmarinic acid and reduced synthesis of analogues. L-amino acid deaminase was introduced to balance metabolic flux, and the stability of caffeoyl-CoA was improved by knocking out the menI gene. The final strain achieved the highest reported production of rosmarinic acid in fed-batch fermentation. The strategies applied in this study provide a foundation for the synthesis of other caffeic acid and rosmarinic acid derivatives.

Ultrasound-assisted extraction of asiaticoside from Centella asiatica using betaine-based natural deep eutectic solvent

Summary: This study developed a novel approach using betaine-based natural deep eutectic solvents (NADES) and ultrasound-assisted extraction (UAE) to extract asiaticoside from Centella asiatica. After screening different NADES, betaine-levulinic acid (Bet-Lev) was found to be the most effective solvent. The extraction process was optimized using response surface methodology (RSM), resulting in an extraction yield of 229.92 +/- 1.67 mg/g. The kinetics of asiaticoside extraction were studied using a second order model, and the proposed extraction mechanisms were supported by morphological analysis. The optimized UAE-NADES technique provided higher yield compared to conventional extraction methods.

Green extraction of phenolic compounds from foxtail millet bran by ultrasonic-assisted deep eutectic solvent extraction: Optimization, comparison and bioactivities

Summary: In this study, ultrasonic-assisted extraction (UAE) using deep eutectic solvent (DES) was applied to extract phenolic compounds from foxtail millet bran (FMB). The optimized extraction process resulted in higher total phenolics, total flavonoids, in vitro antioxidant activity, and acetylcholinesterase inhibitory activity compared to conventional solvent extraction. The phenolic extract from FMB with DES-based UAE contained fifteen phenolic compounds, with p-coumaric acid, apigenin-Cdihexoside, and N', N''-di-p-coumaroylspermidine being the main components.

Effect of solvent type on ultrasound-assisted extraction of antioxidant compounds from Ficaria kochii: Optimization by response surface methodology

Summary: The optimization of extraction conditions for the antioxidant-rich extract from Ficaria Kochii plant was achieved using an ultrasound-assisted system and response surface methodology. The study found that the optimal conditions for extraction using methanol were a solvent-to-raw material ratio of 10%, an extraction time of 50 minutes, and an extraction temperature of 60 degrees C. Under these conditions, the extract had a total phenolic content of 46.379 mg gallic acid equivalents/g dry matter, a total flavonoid content of 11.754 mg quercetin equivalents/g dry matter, and an antioxidant activity of 89.548%. Similarly, the optimal conditions for extraction using water were a solvent-to-raw material ratio of 10%, an extraction time of 30 minutes, and an extraction temperature of 40 degrees C. The resulting extract had a total phenolic content of 51.112 mg gallic acid equivalents/g dry matter, a total flavonoid content of 4.331 mg quercetin equivalents/g dry matter, and an antioxidant activity of 97.004%. This study suggests that the antioxidant-rich extract from Ficaria Kochii could be used as a candidate for food antioxidants and for food storage.

Determination of volatiles, antioxidant activity, and polyphenol content in the postharvest waste of Ocimum basilicum L

Antagonistic storage potential of Tagetes minuta, Eupatorium fortunei and Ocimum basilicum oils with volatile secondary metabolites against Tribolium castaneum and Lasioderma serricorne

Summary: The antagonistic storage potential of Tagetes minuta, Eupatorium fortunei, and Ocimum basilicum volatile oils against Tribolium castaneum and Lasioderma serricorne adults was evaluated. Gas chromatography-mass spectrometry (GC-MS) was used to identify more than 95% of the volatile secondary metabolites obtained at different hydrodistillation times. The results showed that T. minuta oil exhibited the strongest fumigant toxicity against both T. castaneum and L. serricorne. Methylthymol and precocene I showed stronger fumigant and contact toxicity, respectively. T. minuta oil also showed a high repellent rate against both pests. These findings suggest that the essential oils and their main volatile secondary metabolites have the potential to be developed as novel natural botanical resources for antagonistic storage in traditional Chinese medicine (TCM).
