Repurposing FIASMAs against Acid Sphingomyelinase for COVID-19: A Computational Molecular Docking and Dynamic Simulation Approach

Summary: COVID-19 has caused widespread suffering worldwide and research on potential inhibitors against acid sphingomyelinase (ASM) is necessary. Experimental studies show that SARS-CoV-2 activates the acid sphingomyelinase/ceramide pathway, facilitating viral cell entry. This study aimed to inhibit ASM activity to prevent SARS-CoV-2 infection. A drug library containing 257 functional ASM inhibitors was screened using computational molecular docking. Among these inhibitors, Dutasteride, Cepharanthine, and Zafirlukast showed promising results, with low binding affinity scores and non-toxic properties.

Summary: This study aimed to screen for inhibitors of pyoverdine-mediated iron sequestration in P. aeruginosa. Two potential binders, N-2-succinyl ornithine and cilazapril, were found with good drug-likeness and ADME properties. Molecular dynamics studies further confirmed the stability of these complexes. The results suggest that N-2-succinyl ornithine could be explored as a drug for the treatment of P. aeruginosa infections.

Investigating the anti-streptococcal biofilm effect of ssDNA aptamer-silver nanoparticles complex on a titanium-based substrate

Summary: A silver nanoparticle-aptamer complex was designed to remove biofilms on the surface of titanium implants by binding to surface receptors of streptococcal strains. Amyloid Aggregation of Streptococcus mutans Cnm Influences Its Collagen-Binding Activity

Summary: The gene cnm codes for the glycosylated collagen- and laminin-binding surface adhesin Cnm, found in approximately 20% of Streptococcus mutans clinical isolates, associated with systemic infections and increased caries risk. In silico analysis predicted that the N-terminal collagen-binding domain of Cnm has a propensity for amyloid aggregation, while the C-terminal domain was predicted to be disorganized. Thioflavin-T fluorescence and electron microscopy showed that Cnm forms amyloids and the amyloidogenic unit is its collagen-binding domain. Studies characterized the amylogenic properties of Cnm through various assays, indicating its role in recognition, initial binding to collagen-rich surfaces, and potential negative regulation through amyloid formation.
