Application of Marine Natural Products against Alzheimer's Disease: Past, Present and Future

Summary: Alzheimer's disease (AD), a neurodegenerative disease, affects the elderly and manifests as impairments in memory, language, cognition, visuospatial skills, and executive function. Current drugs slow down AD progression and relieve symptoms but cannot cure the disease. The pathogenesis of AD is unclear, but it is associated with beta-amyloid expression. Marine natural products have shown promise as potential drug candidates, with polysaccharides, carotenoids, polyphenols, sterols, and alkaloids presenting neuroprotective effects. Research is ongoing to discover novel and efficacious drug compounds for AD.

Summary: This study developed an in vitro biorelevant methodology to assess the probability of bioequivalence between orally disintegrating tablets (ODTs) taken without water and conventional tablets (CTs) taken with water. The assessment protocol incorporated factors such as the disintegration process in the oral cavity, gastric emptying differences, and fluid shift in the gastrointestinal tract. This methodology can be useful in formulation studies for estimating the bioequivalence probability between ODTs and CTs.

Development and Validation of an HPLC-FLD Method for the Determination of NDMA and NDEA Nitrosamines in Lisinopril Using Pre-Column Denitrosation and Derivatization Procedure

Summary: A new HPLC-FLD method was developed using dansyl chloride for the determination of genotoxic impurities NDMA and NDEA in Lisinopril API and its final product. The method was optimized to achieve high sensitivity and reliable results, and showed linear calibration with satisfactory limit of detection. The proposed procedure is relatively easy, rapid, and suitable for routine analysis tests.

Summary: DiCys and IBLC showed the best overall performance in terms of chiral separation, fluorescence intensity, and ionization efficiency in reversed-phase LC-MS analyses. DiCys/OPA also outperformed Marfey's reagents for chiral separation of amino acids. Overall, DiCys/OPA is an excellent amine-derivatization method for enantiomeric metabolite detection in LC-MS analyses.
