Longitudinal effect of clozapine-associated sedation on motivation in schizophrenia: naturalistic longitudinal study

Noham Wolpe et al.

Summary: Negative symptoms of schizophrenia, such as reduced motivation and impaired emotional expressivity, can be influenced by antipsychotic-induced sedation. This study found that sedation adversely affected motivation but not emotional expressivity in clozapine-treated patients. Clozapine also indirectly impaired motivation through worsening sedation, but after considering sedation, it improved motivation. These findings emphasize the importance of addressing sedative side-effects of antipsychotics for better clinical outcomes.

Avolition as the core negative symptom in schizophrenia: relevance to pharmacological treatment development

Gregory P. Strauss et al.

Summary: Negative symptoms are considered a core component of schizophrenia, with avolition possibly playing a special role among these dimensions. Research indicates that successfully addressing avolition can lead to overall improvement in the constellation of negative symptoms, highlighting its critical importance in treatment.

Apathy in small vessel cerebrovascular disease is associated with deficits in effort-based decision making

Youssuf Saleh et al.

Summary: The study found a relationship between apathy in patients with small vessel cerebrovascular disease and disrupted decision making, as well as abnormalities in the white matter network. Apathetic patients accepted fewer offers on the decision-making task and showed reduced responsiveness to low rewards and aversion to investing in high effort. White matter integrity reductions were specifically associated with apathy, highlighting distinct mechanisms underlying apathy and depression.

Mechanisms Underlying Motivational Dysfunction in Schizophrenia

Youssuf Saleh et al.

Summary: Research on negative symptoms in schizophrenia has provided insights into reward-related processing impairments and dysfunction within specific brain networks. Associations between negative symptoms and reward-processing deficits, cognitive dysfunction, and other factors suggest the importance of objective behavioral and neural measurements in understanding this complex syndrome. Utilizing findings from other brain disorders through transdiagnostic research may shed light on common origins of motivation disorders across diseases and inform future treatment development.

Cognitive control network connectivity differentially disrupted in treatment resistant schizophrenia

Charlotte M. Horne et al.

Summary: Using fMRI and prefrontal cortical glutamate levels, the study investigated how cognitive control differs in treatment-resistant and responsive schizophrenia patients. Differences in effective connectivity within a cortico-striatal-limbic network were found in the patient groups, with resistant patients showing altered connectivity within the ACC. Glutamate levels in the ACC were associated with cognitive control in responsive patients but not in treatment-resistant patients. This study suggests a potential role of aberrant glutamate function in cognitive control impairment in treatment-resistant schizophrenia.
