Bipolar disorder-iPSC derived neural progenitor cells exhibit dysregulation of store-operated Ca2+ entry and accelerated differentiation

Cross-Disorder Analysis of Genic and Regulatory Copy Number Variations in Bipolar Disorder, Schizophrenia, and Autism Spectrum Disorder

Summary: This study aimed to compare the roles of genic and regulatory copy number variations (CNVs) in bipolar disorder (BD), schizophrenia (SCZ), and autism spectrum disorder (ASD). High-resolution CNV data from 8708 Japanese samples were used to perform the largest cross-disorder analysis of genic and regulatory CNVs in these disorders. The results showed differences in CNV burden, characteristics of CNVs associated with neurodevelopmental disorders, and regulatory CNVs between BD and SCZ/ASD. However, there were also shared molecular mechanisms, particularly in chromatin biology. The study identified specific risk genes for BD that could contribute to understanding the pathogenesis of the disorder.

Abnormalities in the migration of neural precursor cells in familial bipolar disorder

Summary: This study investigated the migration patterns of neural precursor cells derived from induced pluripotent stem cells in individuals with bipolar disorder, and found abnormal parameters in cellular migration. Transcriptomic analysis revealed downregulation of genes related to EGF/ERBB proteins. The findings suggest that dysregulation at a collective, systemic level may contribute to the abnormal cellular phenotype in bipolar disorder.

Cellular calcium in bipolar disorder: systematic review and meta-analysis

Summary: The study found an increased basal free intracellular calcium ion concentration in patients with bipolar disorder, even in medication-free individuals. This elevation was observed in mania and bipolar depression, but data was inconclusive for euthymia. Calcium ion response to stimulation with 5-HT or thrombin was also enhanced in cells from bipolar disorder individuals. The results suggest altered calcium functioning in bipolar disorder and call for further research into underlying mechanisms and implications for treatment.
