Assembly of Bleomycin Saccharide-Decorated Spherical Nucleic Acids

Summary: Glyco-decorated spherical nucleic acids (SNAs) are attractive delivery vehicles that can hide the unfavorable distribution properties of loaded oligonucleotides. In this study, glyco-AONARV7 conjugates were successfully synthesized and used to form glyco-decorated SNAs. Cellular uptake experiments demonstrated that these glyco-decorated SNAs efficiently entered prostate cancer cells and showed marked variation in intracellular distribution.

The radiotracer HBED-CC-Tz showed faster and more efficient radiolabeling compared to the DOTA analogue. Both radiotracers demonstrated excellent radiolabel stability in vitro and in vivo.

Immunotherapy for breast cancer using EpCAM aptamer tumor-targeted gene knockdown

Summary: New cancer immunotherapy strategies using EpCAM aptamer-linked small-interfering RNA chimeras (AsiCs) were effective in selectively knocking down genes in EpCAM(+) tumors to enhance immune recognition. Four of the six AsiC (Upf2, Parp1, Cd47, and Mcl1) were able to inhibit tumor growth and improve tumor-infiltrating immune cell functions. AsiC mixtures showed better efficacy than individual AsiC and could synergize with anti-PD-1 checkpoint inhibition.

Summary: A 12-armed Buckminster fullerene with azide functional groups was monosubstituted in organic media with cyclooctyne-modified oligonucleotides, leading to the production of molecularly defined monofunctionalized spherical nucleic acids (SNAs). This controlled synthesis scheme allows for one oligonucleotide arm to be functionalized with labels or conjugate groups, while the other arms can remain unmodified or be decorated with different conjugate groups on the outer sphere of the SNAs. Attention was given to the homogeneity and authenticity of the C60-azide scaffold used for assembling the full-armed SNAs.
