T Cells Promote Metastasis by Regulating Extracellular Matrix Remodeling following Chemotherapy

Jozafina Haj-Shomaly et al.

Summary: Metastasis is a major cause of cancer-related deaths. This study found that chemotherapy can induce a host-mediated mechanism that promotes remodeling of the extracellular matrix (ECM), which facilitates cancer cell seeding and structural changes in the lungs. The research also revealed that the ECM remodeling is mediated by CD8 T cells expressing the enzyme lysyl oxidase (LOX). This study highlights the role of immune cells in regulating the ECM and metastasis following chemotherapy, suggesting that inhibiting chemotherapy-induced ECM remodeling may be a potential therapeutic strategy for metastatic cancer.

Mannosylated poly(acrylic acid)-coated mesoporous silica nanoparticles for anticancer therapy

Haesoo Lee et al.

Summary: In this study, doxorubicin-loaded mesopomus silica nanoparticles (MSNs) coated with mannose grafted poly (acrylic acid) copolymer (DOX@MSNs-man-g-PAA) were developed to enhance the hemocompatibility and target efficacy to cancer cells. The DOX@MSNs-man-g-PAA showed sustained and pH-dependent drug release with improved hemocompatibility and enhanced uptake in breast cancer cells through mannose receptor-mediated endocytosis, leading to increased intracellular ROS and changes in mitochondrial membrane potential. This formulation exhibited superior tumor-suppressing activity in mice inoculated with MDA-MB-231 cells.

Normalizing Tumor Vasculature to Reduce Hypoxia, Enhance Perfusion, and Optimize Therapy Uptake

Kathy Matuszewska et al.

Summary: Solid tumors require rapid development of new blood vessels to support their metabolic needs, which can lead to tumor hypoxia and a pro-tumorigenic environment. Targeting tumor vasculature normalization can reduce hypoxia and enhance therapy efficacy. Understanding tumor angiogenesis biology and exploring new clinical approaches, including combination therapies with immunotherapy, are crucial to advancing vascular targeting as a therapeutic strategy.
