S1P Released by SGPL1-Deficient Astrocytes Enhances Astrocytic ATP Production via S1PR2,4, Thus Keeping Autophagy in Check: Potential Consequences for Brain Health

Shah Alam et al.

Summary: In this study, the neural-targeted ablation of SGPL1 in a mouse model revealed that the accumulation of S1P due to SGPL1 deficiency led to increased expression of glycolytic enzymes, preferential entry of pyruvate into the TCA cycle, and elevated cellular ATP content. The high energy load activated mTOR, thus regulating astrocytic autophagy and potentially affecting neuronal viability.

The role of sphingosine 1-phosphate metabolism in brain health and disease

Gerhild van Echten-Deckert

Summary: Lipids are essential components of the CNS, and sphingolipids, a type of lipid, are highly concentrated in the brain. Sphingosine 1-phosphate (S1P), derived from membrane sphingolipids, has complex effects on the brain, which can be both beneficial and detrimental. However, the role of S1P in brain development and various brain pathologies is still controversial.

Sphingosine 1-phosphate lyase facilitates cancer progression through converting sphingolipids to glycerophospholipids

Baasanjav Uranbileg et al.

Summary: This study reveals the potential role of S1P metabolism and its products in cancer pathogenesis, especially in hepatocellular carcinoma and colorectal cancer. The overexpression of SPL is associated with elevated levels of glyceroLPLs and promotes tumor progression, while the inhibition of SPL has the opposite effect. Furthermore, SPL and glyceroLPLs play important roles in regulating cancer progression through various mechanisms.

The role of Sphingolipids in myelination and myelin stability and their involvement in childhood and adult demyelinating disorders

Paola Giussani et al.

Summary: Sphingolipids play a crucial role in brain development and myelin formation in the nervous system, with abnormalities potentially leading to demyelinating diseases like multiple sclerosis. Understanding the bioactive molecules associated with sphingolipid metabolism, such as ceramide and sphingosine, may help identify potential targets for therapies.
