NLRP3 activation contributes to endothelin-1-induced erectile dysfunction

Summary: In this study, it was hypothesized that stimulation of endothelin receptors ETA and ETB leads to NLRP3 activation through increased calcium and ROS formation. The experiments conducted on mice demonstrated that ET-1 reduced intracavernosal pressure and the CC ACh and SNP-induced relaxation, while increasing caspase-1 expression. The use of receptor antagonists, genetic deletion, and ROS inhibitors reversed these effects, indicating that ET-1-induced erectile dysfunction involves the activation of NLRP3 via Ca2+-dependent ROS generation mediated by ETA- and ETB-receptors.

The findings provide new insights into the mechanism of endothelial cell damage in APS and identify a potential drug target for the clinical treatment of APS.

However, no significant associations were found for IL-1b (3954C/T), IL-2 (330T/G), IL-4 (590C/T), and IL-18 (607C/A) polymorphisms with OLP.

The role of ROS-induced pyroptosis in CVD

Summary: Cardiovascular disease (CVD) is the leading cause of death worldwide and poses a serious threat to human health. Pyroptosis, a newly discovered form of cell death, has been found to play a key role in CVD induced by reactive oxygen species (ROS). However, the exact signaling pathway of ROS-induced pyroptosis is not fully understood. This article reviews the specific mechanisms of ROS-mediated pyroptosis in vascular endothelial cells, macrophages, and cardiomyocytes. Current evidence suggests that ROS-mediated pyroptosis could be a novel target for the prevention and treatment of cardiovascular diseases such as atherosclerosis (AS), myocardial ischemia-reperfusion injury (MIRI), and heart failure (HF).
