Distinct roles for PARP-1 and PARP-2 in c-Myc-driven B-cell lymphoma in mice

Miguel A. Galindo-Campos et al.

Summary: Dysregulation of the c-Myc oncogene is linked to aggressive tumor progression, and PARP-1 and PARP-2 have opposing influence in c-Myc-driven B-cell lymphoma. PARP-1 deficiency accelerates lymphomagenesis, while genetic deletion of PARP-2 prevents B-cell lymphoma. These findings provide important insights for the design of new PARP-centered therapeutic strategies.

miR-149-PARP-2 Signaling Regulates E-cadherin and N-cadherin Expression in the Murine Model of Endometrium Receptivity

Raj Kumar Verma et al.

Summary: E-cadherin is predominantly expressed during the pre-receptive stage of endometrial receptivity and is promoted by PARP-2, which is regulated by miR-149. On the other hand, N-cadherin is only detected at the early stages and decreases thereafter.

PARP inhibitors trap PARP2 and alter the mode of recruitment of PARP2 at DNA damage sites

Xiaohui Lin et al.

Summary: Dual-inhibitors of PARP1 and PARP2 show promise as anti-cancer drugs due to their ability to block enzymatic activity and extend the lifetime of DNA damage-induced PARP1 and PARP2 foci. This study reveals that PARP inhibitors trap PARP2 by physically stalling PARP2 on DNA via the WGR-DNA interaction, while suppressing the rapid exchange of PARP2 dependent on PARP1 and PAR.

Inhibitors of PARP: Number crunching and structure gazing

Johannes Rudolph et al.

Summary: Selective inhibitors of PARP1 and PARP2 are used for treating cancer patients with DNA repair deficiencies. Talazoparib shows higher potency compared to other inhibitors due to its extensive interactions with conserved residues in the active site. The recently characterized protein HPF1 may play a role in designing the next generation PARP1/2 inhibitors.

The BRCT domain of PARP1 binds intact DNA and mediates intrastrand transfer

Johannes Rudolph et al.

Summary: PARP1 is a key player in DNA damage response and a target for cancer treatment. The BRCT domain binds to DNA without activating the catalytic domain. The DNA-binding properties of the BRCT domain contribute to the monkey-bar mechanism for DNA transfer of PARP1.

Summary: PARP 1-3 are multi-domain enzymes known for catalyzing covalent modification of proteins, DNA, and themselves, with important functions in DNA damage response and nucleosome remodelling. Activation of PARP occurs through DNA binding, and modulation of their activity with PARP inhibitors has shown success in cancer therapies.
